To observe the effects of the components a and b of Fomes officinalis Ames polysaccharides(FOPS-a,FOPS-b)on the behavioral changes of APP/PS1 double transgenic mice,and to explore the effects and mechanisms of FOPS-a and FOPS-b on pathological changes in hippocampus.Methods:1)Sixty-four male 3-month-old APP/PS1AD transgenic mice were randomly divided into model group,donepezil group(0.65mg/kg),FOPS-a high,medium,and low(60,30,15 mg/kg)group and FOPS-b high,middle and low(60,30,15 mg/kg)group,wild-type C57BL/6J mice of the same month age were the normal group,8 mice in each group,each group was given drugs and physiological Saline was administered by gavage for 90 days.2)At the age of 6 months,the Morris water maze,diving platform,open field and nesting experiments were used to detect changes in learning and memory ability.3)HE and Nessler staining were used to observe the neuronal morphological changes in the hippocampus of each group;RT-PCR was used to detect the transcription levels of GSK3α,APP,Aβ,AKT,GSK3β,and Tau mRNA in the hippocampus of mice;Western-blot detection of GSK3α,APP,PS-1,Aβ1-42,AKT,GSK3β,Tau,P-tau protein expression.Results:1)In the Morris water maze experiment,the FOPS-a and FOPS-b high(60mg/kg)and medium(30mg/kg)dose groups were significantly shorter in escape latency than the model group(P<0.01),crossing the platform Increased number of times and longer retention time in the target quadrant.The platform jumping,open field and nesting experiments can significantly improve the learning and memory function of mice.2)HE and Nessler staining test results show that the FOPS-a and FOPS-b dose groups significantly reduce neuronal degeneration and edema in the hippocampus compared with the model group,thereby reducing neuronal damage.3)FOPS-a and FOPS-b can up-regulate AKT mRNA transcription level and protein expression,and down-regulate GSK3β,Tau mRNA transcription level and GSK3β,Tau,P-tau protein expression while reducing GSK3α,APP,AβmRNA transcription level(P<0.01,P<0.05)and GSK3α,APP,Aβ1-42,PS-1 protein expression(P<0.01).Conclusion:Fomes officinalis Ames polysaccharide components a and b can improve the learning and memory ability of APP/PS-1 double transgenic mice.By regulating the expression of related proteins in the hippocampus of mice,reducing Aβdeposition and reducing Tau protein hyperphosphorylation,it plays a role in antagonizing AD.
