Objective:To provide a certain experimental basis for further research of polysaccharide,according to the learning of antioxidant,immunomodulatory and anti-tumor activities of polysaccharides from Fomes officinalis Ames,by conducting the experiment in vitro cell and the model experiment in mice.Methods:1、Investigate the In vitro antioxidant capacity of FOP,with the selection of In vitro scavenging free radicals reaction system and the application of DPPH,The development of three phenol from oxidation and Hydroxyl radicals method.Through the employment of MTT Chromogenic method,observe different concentrations of polysaccharides from Fomes officinalis Ames on rat 4TI cell source for breast cancer,colon cancer cell line HT 29,who cervical cancer cell line Hela,human ovarian cancer cell line SK-OV-3,human prostate cancer cells is a PC-3,human liver cell line Hep3 B cell growth inhibition,and sift polysaccharides from Fomes officinalis Ames antitumor activity.3.Use ICR mice to construct the model of 4T1 tumor-burdened mice,observe the influence of polysaccharides from Fomes officinalis Ames on the inhibition of Mice transplanted tumor.4.RSB method is used to investigate the influence of spleen T and B lymphocytes in mice.Discuss the immune function in vitro.Results:1、The result of In vitro antioxidant reveals that polysaccharides from Fomes officinalis Ames are shown in the three kinds of antioxidant system in vitro antioxidant activity.2、In vitro,various dosage strengths of fomes 4Tl of murine breast cancer cell proliferation inhibition.3、Polysaccharides from Fomes officinalis Ames can effectively tumor-burdened solid tumor model mice tumor growth,the results shows that polysaccharides from Fomes officinalis Ames on the inhibitory rate of 4Tl mice were 58.69%(low dose group),60.89%(middle dose group),74.73%(high dose group).4、polysaccharides from Fomes officinalis Ames on spleen cell immune adjustment.Coordination with ConA or LPS FoP can induce mice spleen T and B lymphocyte proliferation,can directly promote the mice spleen T and B lymphocyte proliferation;FoP also can improve the mice spleen cell ratio of CD19 + CD86 + B cells;In addition,the FoP also can promote the secretion of T lymphocytes Th1 cytokine secretion of IL-2 and B cell activation and proliferation of the secretion of IL-13,the main cytokines derived from macrophages TNF-α secretion also has a promoting effect.5、 the FoP can induce the proliferation of peritoneal macrophages in mice,macrophages can cause from the cytokines secreted TNF-α.Conclusion: 1.Polysaccharides from Fomes officinalis Ames has antioxidant activity in vitro.2 Polysaccharides from Fomes officinalis Ames in vivo and inhibit breast cancer cell line murine 4T1 cell proliferation in.3,Polysaccharides from Fomes officinalis Ames can significantly promote the normal mice spleen lymphocyte proliferation,and a dose-response relationship within a certain scope,has immune function.
