Objective:To observe the influence of moxibustion of the‘Eight Methods of Intelligent Turtle' on NF-?B signaling pathway in spleen of aging models of rats,and to explore the mechanism of delaying aging.Methods:SD rats were randomly divided into 6 groups,a blank group,a model group,a prevention group with eight method of intelligent turtle,a prevention group with moxibustion at Shen Que,a group of treatment group with eight methods of intelligent turtleand a treatment group with moxibustion at Shenque(n=10,in eath group).Except the blank group,the other 5 groups were intraperitoneally injected with d-galactose solution to produce subacute aging model.During making model,the prevention groups received the treatments both moxibustion according to opening points in eight methods of intelligent turtle and moxibustion at Shenque for 42 days.After modeling,the treatment groups received the treatments both moxibustion according to opening points in eight methods of intelligent turtle and moxibustion at Shenque for 28 days.The serum levels of TNF-?,IL-1 and IL-6in rats were detected by ELISA.The m RNA expression levels of NF-?B,NF-?B P65 and I?B? in spleen of rats were detected by Real time-PCR.The expression levels of p-P65/P65 protein,p-I?B?/I?B?protein in spleen of rats were detected by Western blot.Result:(1)Compared with the blank group,the expression levels of TNF-?,IL-1and IL-6 in serum of rats in the model group were significantlyincreased(P < 0.01).Compared with the model group,the expression levels of TNF-?,IL-1 and IL-6 in both the prevention groups and the treatment groups were significantly decreased(P < 0.01).There were statistically significant differences in the expression levels of TNF-? and IL-6 in serum between the two prevention groups(P < 0.01,P <0.05).There was no significant difference between the two treatment groups(P>0.05).(2)Compared with the blank group,the m RNA expression levels of NF-?B,NF-?B P65 and I?B? in spleen of rats in the model group were significantly increased(P<0.01).Compared with the model group,the m RNA expression levels of NF-?B,NF-?B P65 and I?B? in spleen of rats in both the prevention groups and the treatment groups were significantly decreased(P<0.05,P<0.01).The m RNA expression levels of NF-?B and NF-?B P65 in spleen of the two prevention groups were statistically significant(P<0.05).There was no significant difference between the two treatment groups(P>0.05).(3)Compared with the blank group,the expression levels of p-P65/P65 protein,p-I?B?/I?B? protein in spleen of rats in the model group were significantly increased(P < 0.01).Compared with the model group,the expression levels of p-P65/P65 protein,p-I?B?/I?B? protein in spleen of rats in both the prevention groups and the treatment groups were significantly decreased(P<0.01).There was no significant difference between the two prevention groups(P>0.05).There was no significant difference between the two treatment groups(P>0.05).Conclusions:(1)Both the moxibustion according to opening points in eight methods of intelligent turtle and moxibustion Shen Que could reduce the expressionlevels of TNF-?,IL-1 and IL-6 in serum.(2)Both the moxibustion according to opening points in eight methods of intelligent turtle and moxibustion Shen Que could reduce the m RNA expression levels of NF-?B,NF-?B P65 and I?B? in spleen,and inhibit the phosphorylation of NF-?B P65 and I?B? protein.(3)It shows that the moxibustion according to opening points in eight methods of intelligent turtle and moxibustion Shen Que have obvious preventive and therapeutic effects on aging rats.The prevention effect of opening points in eight methods of intelligent turtle on aging models of rats may be better than that of moxibustion at Shenque.(4)The mechanism of retarding aging of the eight methods of intelligent turtle may be related to the regulation of NF-?B signaling pathway.
