Objective:Based on previous experiments which confirmed that electro acupuncture at the Foot Shao-yang Meridian has an effect on bone metabolism in castrated SD rats[1],by observing the changes of bone tissue morphology indicators such as parathyroid hormone,PTH,Estradiol,E2,Bone Volume/Total Volume,BV/TV,Trabecular Thickness,Tb.Th,Trabecular Thickness?Tb.Th?,and Trabecular Separation?Tb.Sp?in normal and castrated C57 mice after electro acupuncture at the Foot of Shao-yang Meridian,to further verify the anti-osteoporosis effect of electro acupuncture at the Foot Shao-yang Meridian,to explore the mechanism of acupuncture intervention for postmenopausal osteoporosis,and to provide more scientific theoretical basis for acupuncture to prevent and treat osteoporosis from the two parts of calmodulin and bone tissue morphometry.Methods:1.Animal grouping:40 female C57 SPF mice were randomly divided into Ovariectomized electro acupuncture group,Sham electro acupuncture group,Ovariectomized banding group,and sham operation banding group,10 mice in each group.2.Modeling:Bilateral ovaries were removed from the Ovariectomized removal group,and a segment of fat around the ovary was removed from the sham operation group.3.Intervention method:Electro acupuncture group was treated with electro acupuncture at the Foot of Shao-yang Meridian,GB30,GB39,and GB34.The duration of the intervention was 4 weeks.The sham operated banding group was banded with the same electro acupuncture group,but no electro acupuncture intervention was given.4.Test method:4.1 Serological test:After the intervention,anesthetize the mice,and take 0.5-0.9ml of blood through the inferior vena cava to detect its whole intact-parathyroid hormone?PTH?,Estradiol?E2?.4.2 Test of Bone tissue morphometry:Kill the mice and cut its hind legs,strip the fur and muscles,and fix the complete tibia and femur in paraformaldehyde.Then send them to Southwest Medical University Nuclear Medicine,to analyze the Bone Volume/Total Volume,BV/TV,Bone Surface/Bone Volume,BS/BV,and Trabecular Thickness,Tb.Th,Trabecular Separation?Tb.Sp?and other parameters by the Micro-CT.Results:1.Serological indicators:serum PTH:After intervention,compared with the Ovariectomized electro acupuncture group,the serum PTH of the Sham electro acupuncture group was significantly reduced?P<0.01?.Compared with the sham operation banding group,the PTH in the Sham electro acupuncture group decreased?P<0.05?.Compared with the Ovariectomized banding group,the PTH in the Ovariectomized electro acupuncture group had a decreasing trend,but there was no significant difference?P>0.05?.Compared with the sham operation banding group,E2 in the sham-operated electro acupuncture group had a rising trend,but there was no significant difference?P>0.05?.Compared with the sham operation banding group,the E2 was reduced in the Ovariectomized banding group,but the difference was not statistically significant?P>0.05?.Bone tissue morphometric indicators:After intervention,compared with the Ovariectomized banding group,the BV/TV increased in the Ovariectomized electro acupuncture group?P<0.01?,the BS/BV decreased?P<0.05?,and Tb.Tn increased?P<0.05?,there was no significant difference in Tb.Sp?P>0.05?.Compared with the sham-operated bandage group,the sham-operated electro acupuncture group had increasing tendency in BV/TV and Tb.Tn,and a decrease in BS/BV and Tb.Sp,but the differences were not statistically significant?P>0.05?.Compared with the sham-operated bandage group,the Tb.Sp of the Ovariectomized banding group increased?P<0.05?,the BV/TV decreased?P<0.05?,and the Tb.Tn decreased,but the difference was not statistically significant?P>0.05?,BS/BV increased,but the difference was not statistically significant?P>0.05?.Conclusions:1.Based on the theory of“Shaoyang in charge of bone”,electro acupuncture at the Foot Shao-yang meridian can affect the serum calcium-regulating hormone level in C57 mice.It appears to decrease serum PTH in normal mice and Increase serum E2 in ovariectomized mice.2.Electro acupuncture at the Foot Shao-yang Meridian can improve the micro-bone tissue structure of ovariectomized mice and reverse the changes of bone tissue after ovariectomy.The specific manifestation is the increasing of BV/TV,reducing of BS/BV,and increasing of the thickness of Tb.Tn.3.The anti-osteoporotic effect of electroacupuncture at the points of Foot Shaoyang Meridian on ovariectomized mice may be related to the increasing E2,but not closely related to the PTH.
