Aids:The purpose of this study is to make the model rats of short head tendinitis of biceps,observe the changes of behavior,collagen I and collagen III content and distribution in tendon tissue,and the influence of TGF-β 1,MMP-1 and TIMP-1 content in tendon tissue homogenate,and explore the effect of needle knife intervention on tendon fibrosis of short head tendinitis of biceps And the corresponding mechanism,for the clinical treatment of biceps short head tendinitis needle knife to provide the optimal scheme and data support.Methods:40 rats were randomly divided into normal control group,model group,acupotomotor group and acupotomotor group after adaptive feeding for one week.The rats in model group,acupotomotor group and acupotomotor group were made by collagenase injection combined with excessive exercise method in the first four weeks,and the treatment was started from the fifth week.Rats in the normal control group received routine feeding without any intervention;rats in the model group received routine feeding without any treatment after modeling.After the model establishment of acupotomotor 1 group,the rats were treated with acupotomotor twice a week,once a week: in the first week,the soft tissue under the coracoid process was loosened,that is,the starting point of the tendon of the short head of the biceps brachii muscle;in the second week,the abdominal tenderness of the biceps brachii muscle was loosened;in the second week,the rats in the two groups were treated with acupotomotor twice a week,once a week: in the first and second weeks,the soft tissue under the coracoid process was loosened,that is,the short head of the biceps brachii muscle Tendon starting point.Through open field experiment and direct observation,the behavioral characteristics of rats in each group were analyzed.The changes of type Ⅰ and Ⅲ collagen content and distribution in the tendon of biceps brachii brevis were observed by Sirius red staining.The expression levels of transforming growth factor(TGF-β 1),matrix metalloproteinase-1(MMP-1)and matrix metalloproteinase inhibitor-1(TIMP-1)were detected by ELISA SPSS 22.0 statistical software was used to analyze the experimental data.Results:Behavioral: the results of open field experiment showed that compared with the normal control group,the total activity distance of the model group was significantly shorter(P < 0.01);compared with the model group,the total activity distance of the needle knife group 1 and needle knife group 2 was significantly different(P < 0.01).In the direct observation,compared with the normal control group,the rats in the model group showed morbid state in the manner,posture and hair.Compared with the normal control group,there was no significant difference in the expression,posture and hair between the two groups.2.Histomorphology: in the normal control group,the tendon structure of biceps brachii was clear;the muscle fibers were arranged in parallel and orderly;the membrane of muscle bundles was thin and the collagen fibers were arranged evenly and orderly;the arrangement of collagen fibers was loose and the gap was large;the main collagen was type I collagen with orange yellow color,and a small amount of type III collagen with green color was scattered around type I collagen.In the model rats of short head tendinitis of biceps brachii,the structure of tendon tissue was destroyed,the muscle fibers were twisted,arranged in a disordered and wavy way;the myofasciculus membrane was obviously thickened,the collagen fibers were closely disordered;the arrangement of collagen fibers was disordered,and the space was small;the main collagen was type I collagen with orange yellow color,and a large number of type III collagen with green color was scattered around type I collagen,type III collagen The deposition increased obviously.In group 1,the tendon of biceps brachii muscle was clear and loose as a whole;the muscle fibers were basically arranged in parallel and in order;the membrane of muscle bundle was thin and the collagen fibers were evenly arranged;the arrangement of collagen fibers was loose and the gap was large;the main collagen was type I collagen with orange yellow color,and a small amount of type III collagen with green color was scattered around type I collagen,type III collagen The amount of deposition increased slightly compared with the control group.The tendon structure of biceps brachii muscle in the two groups was clear;the muscle fibers were slightly twisted,small wavy and arranged in order;the arrangement of collagen fibers was disordered;the membrane of muscle bundles was thin,the arrangement of collagen fibers was loose and the gap was large;the main collagen was type I collagen with orange yellow color,the green type III collagen was scattered around type I collagen,and the deposition of type III collagen was found Compared with the normal control group and needle knife group1.3.TGF-β 1,MMP-1,TIMP-1 level: the expression of TGF-β 1,MMP-1,TIMP-1 in tendon tissue of model group was higher than that of normal control group,needle knife 1 group and needle knife 2group,with significant statistical difference(P < 0.01);the expression of TGF-β 1,TIMP-1 in tendon tissue of needle knife1 group was lower than that of needle knife 2 group,and the expression of MMP-1 was higher than that of needle knife 2 group,with significant statistical difference(P < 0.01).Conclusion:1."Collagenase injection combined with excessive exercise" was used to make the rat model of short head tendinitis of biceps brachii,which is feasible and meets the research requirements.2.Acupotomy can significantly improve the shoulder joint activity and motor ability of rats with biceps breviceps tendinitis;3.Needle knife intervention may regulate the expression of myofibrosis related factors in tendons to inhibit the myofibrosis response and reduce the scar area of tendons;4.The therapeutic effect of coracoid process release combined with abdominal tenderness of biceps brachii is better than that of coracoid process release alone.
