Comparison Of The Time Course Effects Of Electroacupuncture At Heart Meridian On The Expression Of DA And 5-HT In Serum And Myocardium Of Rats With Acute Myocardial Ischemia

Objective:To observe the different time course effects of electroacupuncture(EA)at heart meridian on the electrocardiogram(ECG),the concentration of dopamine(DA)and5-hydroxytryptamine(5-HT),and the relative expression of DRD1 mRNA and 5-HT mRNA in myocardial tissue in rats with acute myocardial ischemia(AMI),in order to explore the time-course effect of EA on the myocardial injury of AMI model rats.Methods:110 healthy male SD rats were randomly selected 8 rats as the control group after one week of adaptive feeding,the remaining rats were ligated to reproduce AMI model,the successfully replicated AMI model rats were randomly divided into model group,EA immediate,EA1 d,EA2d,EA 3d,5d,7d and EA 8d,with 12 rats in each group.In addition to the control group and the model group,the AMI rats in each group were treated with EA at different times intervals,at the same time in the afternoon every day,the "Shenmen point" of hand Shaoyin heart channel was selected to be treated with EA at the intensity of 1m A and 2Hz for 15 min.After the last electroacupuncture treatment,8 rats in each group were randomly selected,ECG was collected and recorded,myocardial tissue was cut,the concentration of Da and 5-HT in serum and myocardial tissue was measured by enzyme-linked immunosorbent assay,and the relative expression of DRD1 mRNA and 5-HT mRNA in myocardial tissue was detected by reverse transcription polymerase chain reaction.Result:1)Changes in ST segment: Compared with AMI before replication,the ST segment was raised after AMI replication in each group,which had significantlystatistically significant(P<0.01).Compared with AMI after replication,the model group was not statistically significant(P>0.05),and the ST segment was decreased in EA immediate? 1d group,which had statistically significant(P<0.05),the ST segment was decreased in EA 2d,3d,5d,7d,8d group,which had significantly statistically significant(P<0.01).Changes in T wave: Compared with AMI before replication,the T wave was raised in the model,EA 1d,3d,8d group,which had significantly statistically significant(P<0.01).Compared with AMI after replication,the T wave was decreased in EA 1d,2d,3d,5d,7d,8d group,which had significantly statistically significant(P<0.01).Changes in RR interval: Compared with AMI before replication,the RR interval in each group had some changes,but which was not statistically significant(P>0.05).Compared with AMI after replication,the RR interval after EA at different time intervals had some changes,but which was not statistically significant(P>0.05).Changes in QRS waves: Compared with AMI before replication,the QRS wavelength of each group was changed after AMI replication,but which was not statistically significant(P>0.05).Compared with AMI after replication,the QRS wavelength of the EA at different time intervals had a certain change,and the EA 8d group was significantly statistically significant(P<0.01).Changes in HR: Compared with AMI before replication,the HR of each group was different changed after AMI replication,but which was not statistically significant(P>0.05).Compared with AMI after replication,the HR of EA at different time intervals had a certain change,and the EA 2d group had statistically significant(P<0.05).2)Expression of DA concentration in serum: Compared with the control group,the serum DA in the model group was decreased,which was statistically significant(P<0.05).Compared with the model group,the serum DA in the EA immediate,1d,2d,3d,5d,7d,8d groups was increased,which was significantly statistically significant(P<0.01).Compared with the EA 2d group,the serum DA in the EA 8d group was decreased,which was statistically significant(P<0.05).Expression of DA concentration in myocardial tissue: Compared with the control group,the myocardial tissue DA in the model group was decreased,which was statistically significant(P<0.01).Compared with the model group,the myocardial tissue DA in the EA immediate,1d,2d,3d,5d,7d,8d groups was increased,which was significantly statistically significant(P<0.01).Expression of DRD1 mRNA relative content in myocardial tissue: Compared with the control group,the myocardial tissue DRD1 mRNA in the model group was decreased,which was significantly statistically significant(P<0.01).Compared with the model group,the myocardial tissue DRD1 mRNA in the EA immediate,1d,2d,3d,5d,7d,8d groups were increased,which was significantly statistically significant(P<0.01).3)Expression of 5-HT concentration in serum: Compared with the control group,the serum 5-HT in the model group was increased,which was significantly statistically significant(P<0.01).Compared with the model group,the serum 5-HT in the EA immediate,1d,2d,3d,5d,7d groups was decreased,which was significantly statistically significant(P<0.01).Compared with the EA immediate group,the serum5-HT in the EA 5d,7d,8d grups was increased,which was significantly statistically significant(P<0.01).Compared with the EA 1d group,the serum 5-HT in the EA7 d,8d groups was increased,which was significantly statistically significant(P<0.01).Compared with the EA 2d group,the serum 5-HT in the EA 8d group was increased,which was significantly statistically significant(P<0.01).Compared with the EA 3d group,the serum 5-HT in the EA 5d,7d,8d groups was increased,which was significantly statistically significant(P<0.01).Compared with the EA5 d group,the serum 5-HT in the EA 7d,8d groups was increased,which was statistically significant(P<0.05).Expression of 5-HT concentration in myocardial tissue: Compared with thecontrol group,the myocardial tissue 5-HT in the model group was increased,which was statistically significant(P<0.05).Compared with the model group,the myocardial tissue 5-HT in the EA immediate,1d,2d,3d,5d,7d,8d groups was decreased,but only the EA immediate,1d,2d,3d group was statistically significant(P<0.05).Compared with the EA immediate group,the myocardial tissue 5-HT in the EA 5d,7d was increased,which was significantly statistically significant(P<0.01).Compared with the EA 1d group,the myocardial tissue 5-HT in the EA 7d group was increased,which was significantly statistically significant(P<0.01).Compared with the EA 3d group,the myocardial tissue 5-HT increased in the EA 7d group was increased,which was significantly statistically significant(P<0.01).Expression of 5-HT mRNA relative content in myocardial tissue: Compared with the control group,the myocardial tissue 5-HT mRNA in the model group was increased,which was statistically significant(P<0.05).Compared with the model group,the myocardial tissues 5-HT mRNA in the EA immediate,1d,2d,3d,5d,7d,8d groups was decreased,but only the EA immediate,1d,2d,3d,5d groups was statistically significant(P<0.05).Compared with the EA 1d group,the myocardial tissue 5-HT mRNA in the EA 7d group was increased,which was significantly statistically significant(P<0.01).Compared with the EA 3d group,the myocardial tissue 5-HT mRNA in the EA 7d group was increased,which was statistically significant(P<0.05).Conclusion:1)EA at different time intervals can effectively reduce the ST segment elevation and T wave amplitude of the ECG of AMI model,and for HR,QRS waves and RR intervals in the ECG that there is a certain impact,2)Both DA and 5-HT are involved in the protection of myocardial injury caused by AMI,the EA of the Heart meridian helps to regulate the expression of DA and 5-HT in serum and myocardial tissue,and protects the myocardium;3)The expression of DA and 5-HT in different time course of EA is moreobvious.
