| ObjectiveTo observe the effect of electroacupuncture preconditioning the heart meridian points "Shenmen(HT-7)"-"Tongli(HT-5)" meridian segment on myocardial ischemia reperfusion injury(MIRI)rat electrocardiogram(ECG),fastigial nuclei(FN)neuron firing,field potential,etc.,to explore the influence of electroacupuncture to precondition the anti-MIRI effect of Lateral hypothalamus area(LHA)-cerebellar fastigial nucleus neural loop regulation mechanism.MethodsAccording to the random number table method,male(Sprague Dawley,SD)rats with a body weight of(220±30)g were adaptively fed for one week,and they were divided into sham operation group,model group,electroacupuncture group,damage +electric Needle group,6 in each group.Connect the "Shenmen"(HT7)and "Tongli"(HT5)on the same side to the same pair of positive and negative output lines for electrical acupuncture stimulation,connect to the SDZ-II electronic acupuncture instrument,select the stimulation parameters: the stimulation voltage is 1V,the frequency is 2Hz,and the stimulation time is 20min/time/d,7d in total.According to the coordinates of the LHA(AP: 2.8mm,RL: 1.5mm,H: 8.3-8.5mm)in the damage + electroacupuncture group,a skull drill was used to make a hole after determining the position.Use a micro-injector that has been injected with kainic acid(KA)to slowly insert it into the predetermined site,and slowly inject 0.4μL of 1g/L kainic acid solution on each side at a time.After injection,keep the needle for 5 minutes,and then routinely feed for 3 days.Acupuncture preconditioning intervention was performed,and then the MIRI model was replicated by ligation of the left anterior descending coronary artery.Powerlab multi-channel physiological recorder was used to record the electrocardiogram of each group of rats;the Plexon multi-channel acquisition system was used to record the discharge(Spike,SPK)and field potential(Local Field Potential,LFP)of each group of rat FN neurons.Use Chart5 for Windows software to extract the ST segment displacement value of ECG and compare between groups;Offline Sorter software performs cluster analysis on neuron discharge;Neuro Explorer software performs Auto-correlograms and waveform analysis on neuron discharge(Waveforms),raster analysis(Rasters),frequency histogram analysis(Rate Histograms),spectral energy analysis(Spectrograms),and power spectral density analysis of low-frequency signals in the field potential(Power Spectral Densities),after calculating the local field potential energy by frequency Statistical analysis was performed on the delta band and theta band with differences between the groups.Results1.Compared with the Sham Operation(SO)group,the ST value of the Model(M)group was significantly higher than that of the SO group(P<0.05).Compared with the M group,the ST segment displacement value(P<0.05)of the electroacupuncture(EA)group was significantly reduced.Compared with the EA group,the ST segment displacement value of the lesion+EA(L+EA)group was significantly higher(P<0.05).2.Cluster analysis,autocorrelation analysis,raster analysis,frequency histogram analysis of FN neuron discharge in each group of rats showed that the SO group and the EA group had interneuron discharges with high frequency;the M group and the L+EA group There are pyramidal neurons firing,and the frequency is low.3.The analysis of the spectral energy density of the low frequency signal of the FN local field potential in each group shows that:(1)In terms of the δ band,compared with the SO group,the FN local δ wave of the M group was significantly increased(P<0.01);and the M group.In comparison,the FN local delta wave of rats in the EA group was significantly reduced(P<0.01);compared with the EA group,the FN local delta wave of rats in the L+EA group was significantly increased(P<0.01).(2)As far asθband is concerned,compared with the SO group,the FN localθwave in the M group is significantly increased(P<0.01),and the FN localθwave in the EA group is significantly increased(P<0.01),L+EA The localθwave of FN in group rats increased significantly(P<0.01).4.Correlation analysis between the proportions of local δ waves and θ waves of FN in each group of rats and ST-segment changes shows that the proportions of δwaves are positively correlated with ST-segment changes(P<0.05);the proportions ofθ-waves and ST-segment changes are also positive correlation.(P<0.05).Conclusion1.Electroacupuncture preconditioning the acupoints of the heart meridian can significantly increase the firing frequency of FN inhibitory interneurons in rats with myocardial ischemia-reperfusion injury and reduce the proportion of low-frequency delta wave spectral energy density in local field potentials.2.Electroacupuncture preconditioning the points of the heart meridian can effectively prevent myocardial ischemia-reperfusion injury in rats.This central integration mechanism may be realized through the LHA-FN neural circuit. |
