Objective: ① To probe into etiology and pathogenesis of ischemic heart disease in traditional Chinese medicine and modern medicine , reseach progress in therapeutic effect and its mechanism of acupuncture and moxibustion in theating ischemic heart disease , and application of genechip technique in the field of traditional Chinese medicine and pharmacology. ②To find out heart and brain molecular mechanism of effects of the Heart Meridian of Hand Shao-Yin and the Small Intestine Meridian of Hand Tai-Yang in protecting against acute myocardial ischemia in gene science, so as to clarify the exterior-interior relationship between the Heart Meridian of Hand Shao-Yin and the Small Intestine Meridian of Hand Tai-Yang, furthermore to establish preliminary datebases of 'acupuncture -gene expression pattern' and Look for a group of'specific genes of the relationship between meridians and viscera' . Methods: ①The ancient and modern literature, especially late ten-year' s literature was carefully collected, induced, analysed, generalized and summarized.② Twenty Sprague-Dawley(SD) rats were randomly divided into four grougs, namly model group, Lung Meridian group, Small Intestine Meridian group and Heart Meridian group, each group had five rats. A model of acute myocardial ischemia (AMI) was made by ligation of left anterior descending branch of thecoronary artery (LAD). The meridian sections of 'Shenmen (HT7)-Tongli (HT5)' of the Heart Meridian of Hand Shao-Yin, 'Yanglao (SI5)-Zhizhen(SI7)' of the Small Intestine Meridian of Hand Tai-Yang and 'Taiyuan(LU9)-Lieque(LU7) ' of the Lung Meridian of Hand Tai-Yin were stimulated with multi-electroacupuncture, respectively. A comparative study on changes of gene expression pattern of Lung Meridian group vs model group, Heart Meridian group vs model group and Small Intestine Meridian group vs model group in heart and hypothalamus was done to find common and differential expression genes. Results: Literature research:①Ischemic heart disease was a disease with multiple etiology. The various pathogenic factors could act on different pathologic links, there were many theories in modern medicine. Traditional Chinese medicine emphasized stagnation of heart blood vessels. The disease occurred in the heart, was always connected with the dysfunction of the liver, spleen and kidney. The main pathological characteristics were deficiency in origin and excess in superficiality, the same disease of yin, yang, Qi and blood, and deficiency interwoven with excess. ②Acupuncture and moxibustion had a reliable effect in theating ischemic heart disease, but the therapeutic effects on the all-mentioned were different with the changes of patient' s sex, age, period and condition of illness, and acupoints and method choices as well as efficacy assessment standard. ③After myocardial ischemia , acupuncture and moxibustion could improve cardial disturbance of hemodynamics, recovery myocardial normal metabolism, regulate myocardial contractility and correct arrhythmia throughout central nerve system, cardiaovascular active mass and local myocardial regulation, microcirculation improvement, anti-oxygen free radical, cytokines,NOS and NO and heat shock protein expression regulation. ④Application of Genechip technique was wide and deep with many results in the aspects of essence of TCM syndrome, acupuncture regulation mechanism and Chinese herb effection mechanism, except for the relationship between meridians and viscera. Experimental research: ①Analysis on number ofdifferential expression genes and ESTs in heart and hypothalamus : In heart, Up-and - down-regulated genes and ESTs were 14 with signal log ratio ≥1 and 20 with signal log ratio ≤-1, of 275 and 164 in Lung Meridian group vs model proup, respectively. Up-and-down- regulated genes and ESTs were 20 with signal log ratio ≥1 and 70 with signal log ratio ≤-1, of 329 and 455 in Heart Meridian group vs model proup, respectively. Up-and-down-regulated genes and ESTs were 18 with signal log ratio ≥1 and 26 with signal log ratio ≤-1, of 501 and 347 in Small Intestine Meridian group vs model proup, respectively. In hypothalamus, Up-and-down-regulated genes and ESTs were 57 with signal log ratio ≥1 and 26 with signal log ratio ≤-1, of 553 and 368 in Lung Meridian group vs model proup, respectively. Up-and-down-regulated genes and ESTs were 190 with signal log ratio ≥1 and 34 with signal log ratio ≤-1 , of 1317 and 933 in Heart Meridian group vs model proup, respectively. Up-and-down-regulated genes and ESTs were 122 with signal log ratio ≥1 and 97 with signal log ratio ≤-1, of 1364 and 855 in Small Intestine Meridian group vs model proup, respectively. (2) Analysis on Classification of differential expression genes with signal log ratio ≥1 or ≤-1 in heart and hypothalamus: In heart, differential expression genes with signal log ratio ≥1 or ≤-1 mainly include immumological and inflammatory reaction relative genes,cell signal transduction and transferin relative genes, et al, in Lung Meridian group vs model proup, ion meridian and transport protein relative genes, cell apoptosis and stress reaction protein relative genes, metabolism relative genes, cell signal transduction and transferin relative genes, DNA' s binding, transcription and transcriptional factor relative genes, immumological and inflammatory reaction relative genes, et al, in Heart Meridian group vs model proup , ion meridianand transport protein relative genes, metabolism relative genes, cell signal transduction and transferin relative genes, DNA' s binding, transcription and transcriptional factor relative genes, immumological and inflammatory reaction relative genes, et...
