Objective: To investigate the effect of electroacupuncture(EA) HT7 and HT7 with different matching points on the therapeutic effect of acute myocardial ischemia(AMI) rats;to observe the changes of the Troponin T(c Tn T) in myocardium and serum,and the level of neurotransmitter dopamine(DA) in the hypothalamus area, to compare with the specificity of different matching points in the process of anti myocardial injury, to discuss the central regulatory mechanism of acupuncture on the protection of myocardial ischemia and brain injury, furthermore to research the relationship between meridian, acupoints and the brain, interpretation of "Heart Governing Mind Theory".Methods: Selected 100 healthy male SD rats, placed the microdialysis probe catheters in the rats’ hypothalamus area, leaved the good condition rats, randomly chose 10 to sham-operation group, the rest of the rats were established to AMI model. The AMI model was established by occlusion of the left anterior descending branch of the coronary artery through narcotized by aether; connecting with Powerlab 16 guide physiological recorder,record the electrocardiogram(ECG) and estimated whether the model was successful.Leave the successfully established AMI model rats, and stochastically divided them into five groups: the model group, Shenmen(HT7) group, Shenmen and Xinshu(HT7&BL15)group, Shenmen and Fengfu(HT7&GV16) group, Shenmen and Zhizheng(HT7&SI7)group, 10 rats in each group. All treatments were started on the second day after established model, stimulus current to 1m A, 2Hz, 15 min, once a day for 5 days. After the treatment, the microdialysate was took out immediately, and was detected the level of dopamine(DA) in the hypothalamus area immediately by electrochemical detector. Take the abdominal aortic blood and the myocardial tissue of rats. The content of Troponin T in serum and myocardial tissue was test by ELISA. Data analyzes by SPSS 17.0 for Windows.Results:1.The Powerlab electrocardiogram: Compared with the sham-operation group, the heart rate of the model group was increased,and the ST segment was elevating clearly, the T wave inversion. Display myocardial ischemia model was established successfully.2.The Troponin T was measured by enzyme-linked immunosorbent assay: After the treatment, compared with the sham-operation group, Troponin T in the model group was showed a significant increasing trend, there are have significant difference between these groups(P<0.01); compared with the model group, Troponin T in the treatment group was reduced significantly, there are have statistically significant difference between the groups(P<0.01); compared with the HT7 group, Troponin T in HT7&BL15, HT7&GV16and HT7&SI7 groups were reduced, the differences were statistically significant(P<0.01);compared with the HT7&BL15 group, Troponin T in HT7&SI7 and HT7&GV16 groups were relatively higher(P < 0.05); compared with the HT7&BL15 group, Troponin T in HT7&SI7 group has no significant difference(P>0.05).3. Dopamine(DA) : DA can be measured in each rat hypothalamus area. Compared with the sham-operation group, the level of DA in the model groups were reduced significantly(P<0.01); compared with the model group, DA in each treatment group were decreased significantly(P<0.01); compared with the HT7 group, DA in the HT7&BL15,HT7&GV16 and HT7&SI7 groups were relatively increased; compared with the HT7&GV16 group, DA in HT7&SI7 group has no significant differences(P>0.05);compared with the HT7&BL15 group, DA in HT7&GV16 and HT7&SI7 group were relatively lower(P<0.05).Conclusions:1.EA can obviously change the ECG of AMI rats and promote the heart function.Acupuncture HT7 and BL15 play a significant role in the effect of EA treat AMI, which suggests that acupuncture treatment between yuan-shu points, yuan-source points and xin-du points has a synergistic effect.2.EA could change the dopamine levels in hypothalamus area of AMI rats, which suggests EA have a good therapeutic effect on AMI rats. Compared with using acupoints in heart meridian, acupoints in heart meridian combination with acupoints in du meridian have a better effect on AMI.
