Purpose: To observe the effects of electroacupuncture on the meridians of intestinal flora and its metabolites TMAO and hematological inflammatory factors IL-6 and TNF-a in rats with acute myocardial ischemia, and to explore the "heart meridian-intestinal-heart" action pathway of electroacupuncture on the cardiac meridian to protect myocardial ischemia. Methods: Randomly divided Male SD rats into a sham group,a model group,an electro-acupuncture group of the Heart Meridian(referred to as the Acupuncture Group),a non-menorrhea and non-acupoint group, and a probiotic group, with 6 rats in each group. The left anterior descending coronary artery was ligated to reproduce a model of myocardial ischemia in rats. The acupuncture group used electroacupuncture to stimulate the "Shen Men-Tong Li" segment, with a frequency of 2 Hz and an intensity of 1 m A.Each electroacupuncture stimulation was performed for 30 min once a day for 3 days.In the non-menorrhea and non-acupoint group, electroacupuncture was performed at the non-acupoints of rat hips, the acupuncture time and parameters were the same as those in the acupuncture group. The probiotics group was given a triple probiotic preparation by gavage at a dose of 1.65 m L / 100 g once a day for 3 consecutive days. Before and after copying the model, use the Powerlab 16 guide to observe the changes in the ST segment of the ECG.Enzyme-linked immunosorbent assay(Elisa)for detecting tumor necrosis factor-a(TNF-a),interleukin-6(IL-6)and plasma trimetlylamine oxide(TMAO)content.Variation of the intestinal flora of the small intestine of rats while using 16 S r DNA technology.Results:(1)Compared with the sham group,ST segment elevation of electrocardiogram after modeling is ? 0.1m V;(2)Changes of inflammatory factors and trimethylamine oxide: Compared with the sham group, the levels of IL-6 and serum TNF-a were significantly increased in the model group, and the levels of plasma TMAO were increased significantly also, and the difference reached statistical significance(p <0.01);Compared with the model group,in the acupuncture group and the probiotic group, the levels of TNF-a and IL-6 were significantly reduced, and the plasma TMAO content was significantly reduced(p <0.01);there was no significant change in serum TNF-a,IL-6 content and plasma TMAO content in the non-acupoint and non-acupoint group.(3)Intestinal flora comparison: The intestinal flora abundance in the model group decreased, however,it was not statistically significant;Significant differences in intestinal flora diversity among the five groups(p <0.05);The intestinal flora of the acupuncture group and the probiotic group were clustered with the sham group, while the non-menorrhea and non-acupoint group and the model group were grouped into one group.(4)There is a synergy effect between Psychrobacter and Oligella, and an antagonistic effect between Weeksella and Bifidobacterium.Conclusion:(1)Electro-acupuncture can regulate the intestinal flora and its metabolites in the small intestine of myocardial ischemic rats, thereby inhibiting the expression of inflammatory factors and improving myocardial ischemia;(2)The "Heart Meridian-Intestinal-Heart" action pathway may be one of the effective ways to protect myocardial ischemia by electro-acupuncture.
