Objectives The purpose of this study was to explore the relationship of blood lipids,placental lipid transporters and inflammatory regulators on neonatal weight by detecting and comparing maternal and umbilical blood lipid levels,TLR4 levels in umbilical cord blood and placenta,and the expression levels of FATP2 and FATP4 protein and RNA in placenta,so as to provide new clinical data and theoretical basis for controlling the factors affecting neonatal weight.Methods 1 160 pregnant women with normal pregnancy were collected and labeled as four groups according to the weight of newborns.2 Detection of serum lipid levels in maternal and umbilical vein by automatic biochemical analyzer.3 Detection of TLR4 in umbilical vein blood by double antibody enzyme-linked immunosorbent assay(ELISA).4Detection of TLR4,FATP2 and FATP4 protein expression in human placentas by Western-blot.5 Detection of TLR4,FATP2 and FATP4 mRNA expression in four groups of placentas by RT-PCRT.Results 1 The concentration of TG in maternal elbow venous blood and umbilical venous blood were(2.59±0.91)mmol/L and(1.46±0.35)mmol/L,(2.67±0.60)mmol/L and(1.61±0.29)mmol/L,(2.79±0.74)mmol/L and(1.74±0.28)mmol/L,(3.03±1.06)mmol/L and(1.90±0.40)mmol/L.The levels of TG in maternal and umbilical blood were different by comparing the four groups.The level of TG in macrosomia group increased significantly.The difference have statistical sense(F=4.283,5.957,P<0.05).The HDL in maternal elbow venous blood and umbilical cord blood were(2.44±0.61)mmol/L and(0.82±0.12)mmol/L,(2.19±0.55)mmol/L and(0.76±0.10)mmol/L,(1.91±0.39)mmol/L and(0.72±0.12)mmol/L,(1.88±0.51)and(0.70±0.15)mmol/L.The levels of TG in maternal and umbilical blood were different by comparing the four groups.The HDL level in macrosomia group was significantly lower than that in macrosomia group.The difference have statistical sense(F=5.452,3.142,P<0.05).2 The concentrations of TLR4 in cord blood of the four groups were(3632.37±558.44)pg/ml,(3855.21±774.48)pg/ml,(4089.24±437.63)pg/ml,(4204.38±841.63)pg/ml.The relative values of TLR4 protein and gene in placenta of the four groups were(23.57±5.41 and 136.11±38.02),(30.83±8.89 and 162.45±36.01),(47.79±10.68 and 183.35±52.28),(48.90±10.94 and 202.90±42.77).Neonatal weight gain with the increase of TLR4 level in umbilical cord blood and placenta.The difference have statistical sense(F=1.930,25.814,8.766,P<0.05).3 The relative values of FATP2 protein and FATP2 mRNA in placenta of the four groups were(101.84±10.64 and 81.21±25.71),(138.55±23.31 and 105.75±37.09),(180.90±31.08 and 113.70±41.73),(229.60±26.50 and146.65±51.86).Neonatal weight gain with the increase of the expression of FATP2 protein and gene in placenta.The difference have statistical sense(F=101.095,8.731,P<0.05).4The relative gray levels of FATP4 protein and gene in placenta were(128.79±66.03 and93.89±51.05),(157.60±56.70 and 112.10±50.25),(169.65±57.68 and 138.15±53.95),(195.65±47.93 and 151.80±67.67).Neonatal weight gain with the increase of the expression of FATP4 protein and gene in placenta.The difference have statistical sense(F=4.567,4.163,P<0.05).5 Logistic regression analysis showed that the expression levels of the weight of placenta,the expression levels of TLR4,FATP2 and FATP4 in placenta were all related to neonatal weight.The expression level of FATP2 increased,and neonatal weight gain was more significant(OR=4.824),with signifi-cant difference(P< 0.05).Conclusions 1 There were differences in blood lipid level and umbilical vein blood lipidlevel among pregnant women with different neonatal weight.The level of TG in maternal blood and umbilical cord blood of gigantic infants increased significantly,while the level of HDL decreased significantly.2.The higher the expression level of TLR4 in cord blood and human placenta,the greater the weight of newborns.3 The expression levels of FATP2 and FATP4 in human placenta increased,and neonatal weight increased.The expression level of FATP2 increased,and neonatal weight increased more significantly.Figure6;Table15;Reference 94...
