Objectives In this study,the serum lipid levels of maternal blood and umbilical venous blood,the level of TLR4 in umbilical blood and the expression levels of TLR4,LPL and FAT/CD36 in placental tissue of the two groups of pregnant women were detected,and the relationship between blood lipid,inflammatory factors and lipid transporter with GDM were analyzed.Meanwhile,the relationship between blood lipid,TLR4,LPL and FAT/CD36 and neonatal weight in GDM pregnant women was also discussed,so as to provide theoretical basis for the influence of GDM pregnant women on neonatal weight.Methods Fifty GDM pregnant women with satisfactory blood glucose control were collected as the case group,and 50 pregnant women with normal glucose tolerance in concurrent delivery were collected as the control group.Venous blood of the fasting elbow of the two groups of pregnant women was extracted 2 weeks before delivery,and venous blood of the umbilical cord of the two groups of newborns was extracted during delivery.The maternal blood and cord blood levels of TG,TC,LDL-C and HDL-C were respectively tested in the laboratory department of our hospital;TLR4 levels in serum of umbilical veins were detected by double antibody sandwich enzyme linked immunosorbent assay(ELISA);Western blot analysis of TLR4,LPL and FAT/CD36 protein expression in placental tissue;RT-PCR analysis of TLR4,LPL and FAT/CD36 mRNA expression in placental tissue.Collate data and statistical analysis.Results 1 There was no statistically significant difference in general information and HbA1 c between the two groups(P>0.05),and the data of the two groups were comparable Conclusions;2 The maternal blood TG level(3.30±1.04)mmol/mL in the case group was higher than that in the control group(2.79±1.07)mmol/mL,and the maternal blood HDL-C level(1.75±0.31)mmol/mL in the case group was lower than that in the control group(2.10±0.41)mmol/mL,with statistically significant differences(t=2.430,-4.777,P<0.050).Maternal blood TC and LDL-C levels were not statistically significant between the two groups(P>0.05).There were no significant differences in TG,TC,LDL-C and HDL-C levels between the two groups(P>0.05).The TG level in maternal blood of the case group was positively correlated(r=0.815,P<0.05),and negatively correlated with hdl-c level in maternal blood(r=-0.683,P<0.05).while no correlation was found with maternal blood TC,LDL-C,umbilical cord blood TG,TC,LDL-C and HDL-C levels(P>0.05);3 The serum TLR4 level of umbilical venous blood(4193.02±799.53)pg/mL in the case group was higher than that in the control group(3669.26±769.65)pg/mL,and the difference was statistically significant(t=3.337,P<0.05).The relative expression of TLR4 protein and mRNA in placenta of the case group(0.85±0.116,0.86±0.099)was higher than that of the control group(0.77±0.117,0.80±0.123),and the difference was statistically significant(t=3.347,2.821,P< 0.05).Neonatal body weight in the case group was positively correlated with the level of TLR4 in umbilical vein blood(r=0.887,P<0.05),and the relative expression of TLR4 protein and mRNA in placental tissue(r=0.869,0.858,P<0.05);4 The relative expression of LPL protein and mRNA in placenta in the case group(0.73±0.141,0.71±0.121)was lower than that of the control group(0.79±0.110,0.79±0.114),and the difference was statistically significant(t=-2.411,-3.184,P<0.05).No correlation was found between neonatal weight and the relative expression of LPL protein and mRNA in placental tissues in the case group(r=-0.150,0.159,P<0.05);5 The relative expression of FAT/CD36 protein and mRNA in placenta of the case group(0.85±0.130,0.83±0.114)was higher thanthat of the control group(0.76±0.113,0.76±0.114),and the difference was statistically significant(t=3.79,2.814,P<0.05).Neonatal body weight in the case group was positively correlated with the relative expression of FAT/CD36 protein and mRNA in placental tissue(r=0.885,0.831,P<0.05).Conclusions 1 Maternal blood TG and HDL-C in GDM pregnant women were significantly increased in the third trimester;The higher the TG level,the lower the hdl-c level,and the higher the neonatal weight;2 The expression level of TLR4 in umbilical venous blood and placental tissue of GDM pregnant women was increased.The higher the expression level,the higher the newborn weight.3 LPL expression level was decreased in the placental tissue of GDM.However,no relationship between the expression level and neonatal weight was found.4 The expression level of FAT/CD36 in the placenta of GDM was increased.The higher the expression level,the higher the newborn weight.Figure 6;Table14;Reference 135.
