OBJECTIVE: Allergic rhinitis(AR)is a non-infectious chronic inflammatory disease of the nasal mucosa that is mainly mediated by Ig E after exposure to allergens.The incidence of AR is related to genetic,environmental,ethnic and other risk factors.This article uses meta-analysis to study whether TNF-? gene 308A/G polymorphism is associated with susceptibility to allergic rhinitis.The relationship between AR susceptibility and genetic mechanism was analyzed in order to provide a theoretical basis for the pathogenesis of AR and disease prevention from a genetic perspective.Methods: The literatures on TNF-?-308A/G polymorphism and asthma susceptibility were collected by searching Pub Med,Web of Science,China Knowledge Network,Wanfang database and other databases.Rev Man5.3 software was used for Meta.analysis.RESULTS: According to the exclusion criteria,a total of 5 articles were included in the study.A total of 1799 subjects were included,with a total of 912 cases in the case group and 887 cases in the control group.The results of meta-analysis showed that the genotype distribution of the case group and the control group were statistically significant.The genetic models were recessive gene model(AA vs GG+GA)and codominant gene model(AA vs GG).The combined OR of the genetic model(AA vs GG+GA)was 2.91,95% CI was [1.44,5.88],P = 0.003,indicating that the TNF-? gene 308G/A site carries a homozygous recessive mutation genotype AA with AR The risk was 2.91 times higher than that of wild-type homozygous GG and heterozygous mutation GA.The co-dominant gene model(AA vs GG)had a combined OR of 3.89,95% CI [1.92,7.89],P = 0.0002),indicating that the TNF-? gene 308G/A site carries a homozygous recessive mutant gene.The risk of AR in type A A was 3.89 times higher than that in wild-type homozygous GG.There were no statistically significant differences in genotype distribution between the case group and the control group.The gene models were dominant gene model(GA+AA vs GG)and superdominant gene model(GG+AA vs GA).Dominant gene model(GA+AA vs GG)with a combined OR of 1.59,95% CI [0.87,2.89],P = 0.13,superdominant gene model(GG+AA vs GA),combined OR value of 0.70,95 %CI is [0.42,1.15] and P = 0.16.There was no significant difference in the allele frequency distribution between the case group and the control group.The allele model(A vs G)had a combined OR of 1.54,95% CI [0.94,2.53],P = 0.09.CONCLUSION: TNF-? gene 308G/A site carries a homozygous recessive mutant genotype AA,which significantly increases AR susceptibility,while TNF-? gene 308G/A site does not increase AR when allele A mutation occurs.Susceptibility.
