Objective: Allergic rhinitis (AR) has a complex genetic background. It is generally considered to be caused by the interaction of genetic and environmental factors. The genetic basis of allergic rhinitis is still a matter of intensive research and efforts have been expended in mapping the chromosomal locations of the susceptibility genes. The aim of our study is to study on the association between the gene polymorphism of interleukin-4(IL-4) and interleukin-4receptor (IL-4R) with the allergic rhinitis in Uygur, thereby exploring the candidate genes and providing information for the prevention and treatment of Allergic rhinitis.Methods: Using community-based case-control study to explore the 109 Uygur patients of allergic rhinitis (case group) and 393 healthy people (control group), both of them living in Kashi area of Xinjiang. The IL-4R rs6498011A/G and IL-4 rs2243283 C/G polymorphism were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), comparing the distribution frequency of genotype and allele between the two groups .The relevant factors of AR were analyzed by questionnaire investigation. Results: (1) The distribution frequency of genotypes of IL-4R rs6498011A/G has significant difference between case and control groups. (x 2=23.841,P<0.001), The distribution frequency of alleles has significant difference too (x 2=7.113 , P=0.008) .Logistic regression analysis equally showed that the IL-4R rs6498011 genotypes closely related with allergic rhinitis. The risk of suffering allergic rhinitis among those with genotypes AA and AG was 7.330 and 5.316times in comparison with those with genotypes GG , respectively. (2)The distribution frequency of genotypes and alleles of IL-4 rs2243283C/G has no significant difference between case and control groups(P>0.05). (3) Smoking are closely related with allergic rhinitis in Uygur, living in Kashi area of Xinjiang. The risk of suffering allergic rhinitis among smokers was 2.073times in comparison with non smokers.Conclusion: The data suggests that the gene polymorphism of IL-4R rs6498011A/G was strongly associated with allergic rhinitis in Uygur and it is might be one of the candidate genes for allergic rhinitis in Uygur; There are no significant association between the gene polymorphism of IL-4 rs2243283 C/G and allergic rhinitis in Uygur.
