ObjectiveTo investigate the effects of Mongolian medicine thirteen-flavored Honghua Miaowan pill on the expression of related factors in miR-146 a,miR-155 and TLR/NF-?B signaling pathways in thymus of rats with allergic rhinitis.The role of Mikuwan in the treatment of allergic rhinitis and its related mechanism of action,to observe whether this treatment is effective for the treatment of allergic rhinitis,and to provide experimental basis for the wider application of the drug in clinical.MethodsIn the experiment,sixty rats were randomly divided into 6 groups,with 10 rats in each group: high-dose Mongolian medicine group,medium-dose Mongolian medicine group,low-dose Mongolian medicine group,Qianbai rhinitis group,blank control group and model control group.AR rat model was established by intraperitoneal injection of ovalbumin and local nasal mucosa challenge.The rat behavioral score and PCA passive skin allergen test were used to verify whether the model was successful.After the modeling,each group was given the corresponding drug treatment(except the blank group).After four weeks of continuous intervention,the rat thymus tissue was taken out and the expression of miR-146 a,miR-155,TLR4,NF-?Bp65 in the thymus tissue were detected by RT-PCR.Results(1)Except the blank group,the behavioral scores of rats in each group were greater than 5 points,and higher than the blank group(p<0.05),subjectively indicating successful modeling.The results of PCA showed that the results of 10 ratsin the blank group were negative,and the diameter of the back was less than 5 mm.Among the 10 rats in the model group,8 were 10 mm,2 were between 5 and 8 mm,and the rats had positive reactions.It was proved that the OVA antibody was present in the rat,which objectively showed that the basic model was successful.(2)The expression levels of miR-146 a,miR-155,TLR4 and NF-?Bp65 in thymus tissues:Compared with the model group,the expression of miR-146 a in the high,middle and low dose groups of Mongolian medicine increased,miR-155.The expression levels of TLR4 and NF-?Bp65 were all decreased.Among them,the Mongolian medicine high-dose group and the Qianbai rhinitis group were effective,and the Mongolian medicine in the low-dose group was not as effective as the Mongolian high-dose group and the Qianbai rhinitis group.Conclusions(1)High,medium and low doses of Mongolian medicine have a certain effect on allergic rhinitis,and the high dose group has the best effect.(2)One of the possible mechanisms of the treatment of allergic rhinitis by thirteen-flavored red flower capsules is to reduce the expression of miR-145 a and reduce the expression of miR-155,thereby inhibiting the key factor TLR4 in the TLR/NF-?B signaling pathway.The expression of NF-?Bp65 is released,thereby improving the inflammatory damage of allergic rhinitis and treating allergic rhinitis.
