Objective:The effects of nuclear case injection combined with Mengyao Rilun Pill on allergic rhinitis(AR)and AR obese rats were investigated.The mechanism of amino acid changes on AR and AR obese rats was found.The theoretical basis was provided for the clinical treatment of AR obese patients with nuclear case injection combined with Mengyao Rilun Pill.Methods:1.Seventy-two male Wistar rats of SPF grade were randomly divided into nine groups.Group A was a blank group,group B was a model group,and group C was a triamcinolone injection group.Groups D,E,and F were AR nucleus.Casein injection combined with Mongolian medicine Rilun Pill high,medium and low dose intervention group;G,H,I groups were AR obese type administration of nuclear case injection combined with Mongolian medicine Rilun Pill high,medium and low dose intervention group.The A-F group was fed with conventional rat feed,and the G-I group was fed with high-fat rat feed to establish an obese rat model.2.B-I groups were prepared by intraperitoneal injection of ovalbumin and nasal mucosal stimulation.3.nasal mucosal tissue was HE stained in AR group and obese group after4.weeks of drug intervention to observe the morphological changes and cell pathological changes.4.blood was collected and the changes of plasma HIS?Ig E?IL-4?IL-6 in each group were detected by ELISA method.5.use liquid-mass spectrometry(LC-MS/MS)metabolic histology to study the changes of 22 amino acids in plasma of rats in each group,and to analyze the mechanism of drug action and the effect of amino acids on disease.Results:1.AR modeling was successful,and the nasal symptoms of each group were scored.There was a significant difference in the cumulative score of nasal symptoms in group B-I compared with group A(P<0.01).Obesity modeling was successful.The body weight of each mouse and the number of Lee's indicators were calculated.There was a significant difference between the G-I group rats and the A-F group rats(P<0.01).After four weeks of drug intervention,there was a significant difference in nasal symptom scores between group B and the other groups(P<0.01);and the weight of rats in group G-I decreased,indicating that Rilun Pills had the effect of reducing obesity.2.Groups A,C,and G have similar morphological changes in the nasal mucosa of rats;nasal mucosa shedding and inflammatory cell infiltration can be found in group D,and inflammatory cell infiltration can be found in comparison with group B Significantly reduced;the degree of nasal mucosal destruction and swelling were more severe in the EF and HI groups,with varying degrees of bleeding and infiltration of inflammatory cells.Among them,the I organization was the closest to group B.3.Elisa results in rat plasma showed that compared with group A,the levels of HIS,Ig E,IL-4,and IL-6 in group B plasma were significantly increased(P<0.05).After drug intervention,the levels of detection factors in the plasma of all groups were reduced.There was no difference between group B and group F and I(P>0.05),and there was no difference between group G and H and group C(P> 0.05).4.Metabolomics-targeted amino acid experimental study: Ala,Ser,Orn,His,and Arg contents in AR are up-regulated by comparing amino acid contents.Pro,Val,Thr,Ile,Leu,Asp,Gln,Lys,Glu,Lys,The contents of Met,Phe,Tyr,and Trp showed a decreasing trend;there was no difference between group F,I and group B after drug intervention(P>0.05),and there was a statistical difference between groups C,D,E,G,and H and group B Significance(P<0.05).Multivariate statistical analysis showed that groups D and G tended to group A,of which group G was more significant;Ala,Arg,Lys,Tyr,Pro,Thr,Leu,and Ile were identified as potential biomarkers for AR.Conclusion:1.Nuclear case injection combined with Mongolian medicine Rilun Pill can effectively improve the typical symptoms and nasal mucosa of AR and AR obese rats,reduce the content of Ig E,HIS,IL-4,IL-6 in plasma and affect obese AR More significant.2.Metabonomics studies have shown that nuclear casein injection combined with Mongolian medicine Rilun Pill significantly regulates AR obese metabolites,and changes in amino acid content have effects on AR and AR obese rats.
