Objective: to establish a model mouse model of lung-qi deficiency by cigarette smoking combined with Senna leaf perfusion,and to observe the weight change of mice before and after treatment with Shenqi granule,and compared with Yupingfeng granule,and compared with that of Yupingfeng granule,and compared it with Yupingfeng granule.The changes of spleen/ thymus organ index and serum TNF-? and IL-1? levels in mice provideexperim ental and theoretical basis for the treatment of recurrent respiratory tract infection with Shenqi granule.Methods: 60 Kunming(KM)mice of clean grade were selected and fed adaptively for 3 days under the same conditions.The mice had no adverse reaction,eating,drinking water and normal activity,and were confirmed to be healthy mice.The mice were randomly divided into 5 groups,12 in each group,namely,blank control group,model group,Yupingfeng granule group,Shenqi granule low-dose group and Shenqi granule high-dose group.With the exception of blank control group,the model of lung-qi deficiency was induced by cigarette smoking combined with Senna leaf perfusion for 10 days in the remaining group.Group A(blank control group): normal saline(NS)group(control group)was given intragastric administration of 0.2ml twice a day for a total of 20 days after successful establishment of the model.Group B(model group): normal saline;group C(Yupingfeng granule group): 4.09g/Kg times;group D(Shenqi granule low dose group): 13.36g/Kg times;group E(Shenqi granule high dosage group): 26.73g/Kg times.After the last dose of medicine,fasting can not help the water.Chloral hydrate was anesthetized by intraperitoneal injection of 0.3ml/100 g the next day,one eyeball was removed and 5ml of blood was taken from each mouse.After 20 minutes incubation in 37? water bath pot,the blood vessels were removed and put into centrifuge,centrifuged at 2500 rmin,centrifuged for 15 minutes,and the supernatant was divided into 2 000 r/min and 15 min respectively.The centrifuge tube of 0.5ml was placed in the refrigerator at-80?.Serum freezing was used to detect the contents of TNF-? and IL-1?.The spleen of the dissected mice was dissected and the thymus was weighed by electronic balance.The organ indexes of the spleen and thymus of each group were calculated after the experiment.All the data were expressed as mean ±standard deviation(`x±s),and the comparison between groups was analyzed by one-way ANOVA.All data were processed by SPSS19.0 statistical software.Results:(1)the weight of mice in the blank group showed a steady increasing trend,compared with the blank group,the weight of the mice in the model group was reduced after modeling,the difference was statistically significant(P<0.05),and the body weight of the mice in the model group was significantly lower than that in the blank group(P< 0.05).After treatment,the body weight of the model group was not significantly increased,but the weight of the Yupingfeng group,high Shenqi granule group and low dose group showed an increasing trend,but there was no significant difference between the groups(P>0.05).(2)Spleen/thymus index: the index of spleen and thymus in the model group was significantly lower than that in the blank control group(P<0.01),and the difference was statistically significant.The spleen index and thymus index of the mice treated with Yupingfeng granule and Shenqi granule(high and low dose)were significantly higher than those of the model group(P<0.01).The dose-effect relationship between Yupingfeng granule group and Shenqi granule group(high and low dose)was not statistically significant(P>0.05),and there was no significant difference between Yupingfeng granule group and Shenqi granule group(P>0.05).(3)Changes of TNF-? and IL-1? in mice: compared with the blank control group,the model group was higher than the control group,and the difference was statistically significant(P<0.05).Compared with the model group,the drug treatment groups were lower than the model group,the difference was statistically significant(P<0.05).Compared with Yupingfeng group,Shenqi granule was lower than Yupingfeng group,and high dose group was lower than Yupingfeng group,the difference was statistically significant(P<0.05).Compared with the low dosage group of Shenqi granule,the high dosage group of Shenqi granule was significantly lower than that of the low dosage group of Shenqi granule,the difference was statistically significant(P<0.05).Conclusion: 1.Shenqi granule can obviously increase spleen and thymus index,decrease the contents of TNF-? and IL-1?,inhibit the inflammatory cascade reaction triggered by these factors,and improve the immune function of the body.2.The effect of Shenqi granule on the treatment of recurrent respiratory tract infection with lung-qi deficiency is superior to that of jade screen granule.
