Clinical Significance Of Expression Levels Of MiR-21-5p,miR-590-5p And MiR-675-5p In Serum Of Patients With Gastric Cancer

Objective : To detect the expression levels of miR-21-5p,miR-590-5p and miR-675-5p in the serum of patients with gastric cancer before and after operation and healthy people,and to explore the relationship between miR-21-5p,miR-590-5p and miR-675-5p and clinical pathology of patients with gastric cancer,and their clinical value in early diagnosis and prognosis assessment of gastric cancer.Methods : 1.RT-PCR was used to detect the relative expression of miR-21-5p,miR-590-5p and miR-675-5p in serum of 50 patients with gastric cancer(32 patients were matched preoperatively and postoperatively)and 30 healthy persons.The expression of miR-21-5p,miR-590-5p and miR-675-5p in serum of pre-operation,post-operation and healthy persons were compared.2.To analyze the relationship between the relative expressions of miR-21-5p,miR-590-5p and miR-675-5p in serum and the clinicopathological features of gastric cancer.3.ROC curves were drawn to evaluate the diagnostic value of miR-21-5p,miR-590-5p and miR-675-5p in gastric cancer.Results: 1.The expression levels of miR-21-5p in serum of patients with gastric cancer were higher than those of healthy controls(P<0.05),while the expression levels of miR-590-5p and miR-675-5p were lower than those of healthy controls(P<0.05).The expression levels of miR-21-5p in serum of patients with gastric cancer before operation were higher than those after operation(P<0.05),while the expression levels of miR-590-5p and miR-675-5p in serum of patients with gastric cancer after operation were higher than those after operation(P<0.05).2.The expression of miR-21-5p was negatively correlated with the differentiation degree of gastric cancer(P<0.05),but not with the age,gender,TNM stage,tumor size and lymph node metastasis(P>0.05).The expression of miR-590-5p was negatively correlated with lymph node metastasis of gastric cancer(P<0.05).No correlation was found between TNM stage,tumor size and differentiation degree(P>0.05).No correlation was found between miR-675-5p expression and age,gender,TNM stage,tumor size,differentiation degree and lymph node metastasis(P>0.05).3.ROC curve analysis showed that the AUC of serum miR-21-5p,miR-590-5p and miR-675-5p were 0.762(95% CI: 0.658 ~ 0.866,P<0.05).Conclusion: 1.miR-21-5p is highly expressed in the serum of patients with gastric cancer and is associated with differentiation of gastric cancer.MiR-590-5p is lower expressed in the serum of patients with gastric cancer and is associated with lymph node metastasis in gastric cancer.2.The expression levels of miR-21-5p,miR-590-5p and miR-675-5p in serum of patients with gastric cancer were different before and after operation.
