Objective: To detect the abundance of microRNA-100(miR-100) in gastric cancertissues(GCTs) and normal adjacent tumor tissues (NATs), and that in the serum ofpatients with gastric cancer (GC) and with healthy control; comparison the expressionof miR-100in gastric cancer tissue and normal adjacent tumor tissues,also in theserum of patients with gastric cancer (GC) and with healthy control,whose relevanceto clinicopathological characteristics was further investigated, in order to observe theclinical significance of miR-100as a serum marker for cancer detection.Methods: Taqman stem-loop real-time RT-PCR method was used to assess theabundance of miR-100in the serum (including40GCs and40healthy controls) andthat in the tissue (40resectable cases of GCTs and NATs); The pathologic and clinicaldata of the patients with gastric cancer and the clinical data of healthy controls werealso collected. The relationship of the miR-100expression in gastric cancer tissuesand normal adjacent tumor tissues was analyzed, gatric cancer patients and healthyserum miR-100differences, and the relationship of the miR-100expression in gastriccancer tissues and serum and clinical indexes were also analyzed.Results:(1) The expression level of miR-100was significantly lower in theNATs than that in the paired GCTs (P<0.05). No significant difference of miR-100level was observed between clinicopathological characteristics (different clinicalstages,different lymph node metastasisstages or different tumor stage).(2) MiR-100was over expressed in the serum of gastric cancer compaired with thatin the healthy control(P<0.05).The stomach cancer patients serum, the expression ofmiR-100in patients with low differentiation higher than the patients with highdifferentiation, but not statistically significant.Further analysis found that miR-100indifferent number of lymph node metastasis and invasion depth and expression in the staging of gastric cancer patients also no significant difference (P>0.05).(3) The ROC curve analysis found that miR-100, CEA,AFP, CA153, CA199andCA125area under the curve were0.985,0.814,0.795,0.744,0.729,0.721. The areaunder the curve of miR-100up to0.985, indicating that the use of serum expressionof miR-100in the diagnosis of gastric cancer have a higher accuracy. Furthercomparative analysis found that miR-100in0.48fmol/l critical value for the diagnosisof gastric cancer, the sensitivity was95%and specificity of97.5%.Coqnclusion:(1) High expression of the miR-100in gastric cancer shows that it mayact as a tumor promoting in the pathgenesis of gastric cancers.(2)The serum levels of miR-100in gastric cancer were over expressed than that in thehealthy control and statistical learning differences; expression of miR-100in serum ofgastric cancer patients with clinicopathological characteristics did not correlation.(3) The area under ROC curve reached0.985, indicating that the expression of serummiR-100for the diagnosis of gastric cancer have higher accuracy. Gastric cancerpatients with miR-100sensitivity and specificity were higher than CEA,AFP, CA153,CA199and CA125in tumor.MiR-100can be used as indexes of gastric cancer.
