In recent years,the research on the properties of traditional Chinese medicine has made remarkable achievements in chemical basis and pharmacodynamics.However,most of them take single herb as the object of study,and fail to take into account the interaction between different stir-frying products and other herbs in the compatibility of compound prescriptions[1-2].Because of the complexity of the change of material base before and after blending,the study of compound prescription,the change of substance basis before and after compatibility,and the change of pharmacodynamics are more consistent with the characteristics of clinical use of traditional Chinese medicine.Linggan Wuwei JiangXin decoction,contained in the synopsis of the Golden Chamber,is a classic prescription for clinical treatment of cold drink and lung,and with dry ginger as the monarch.Ginger grows in resolving the surface of the cold,stop vomiting and phlegm,dry ginger warm in the cold,back Yang Tongmai,stir-frying ginger can warm meridian hemostasis,temperature to relieve pain[3].What is the basis for the selection of ginger gun products in Linggan Wuwei JiangXin decoction?What is the effect of the compatibility of different ginger gun products on its material basis and action mechanism?It is an important scientific problem to clarify the material basis and mechanism of effect of ginger before and after processing by means of technical means in accordance with the theory and thinking of traditional Chinese medicine.ObjectiveOn the basis of the previous study of our research group,the chemical constituents of Linggan wuwei JiangXin?fresh/dry/stir-frying?were investigated in order to identify the chemical constituents contained in Linggan wuwei JiangXin decoction?fresh/dry/stir-frying?.To establish cold-drink-lung asthma model,analyze and compare Linggan wuwei JiangXin decoction?fresh/dry/stir-frying?on the substance and energy metabolism index of cold asthma rats,and find out the effective substances in treating cold disease asthma by combining the correlation analysis of spectrum effect.It provides experimental basis for processing and compatibility study of ginger.Method1.Optimization of decoction of Linggan Wuwei Jiangxin DecoctionThe content of total phenolic acid?total phenolic acid?of Glycyrrhiza uralensis,the single-salt of glycyrrhizic acid,6-gingerol,schizandrin,asarum,extract and total phenolic acid were studied.A multi-index orthogonal test and a comprehensive weighting method were used to carry out the statistical analysis on the test results,Finally,the water decoction technology of Linggan Wuwei Jiang Xin Tang was optimized,which provided the experimental basis for the preparation of Linggan Wuwei Jiang Xin Tang sample?fresh/dry/stir-frying?.2.The effect of fresh,dry and stir-frying ginger combined with Linggan Wuwei Jiang Xin decoction on the chemical constituents of the whole prescriptionAccording to the water decoction process that has been optimized in the previous period,10 batches of fresh,dry and candied glutinous Wuwei Jiangxin soup were prepared for the test samples.Ten batches of Linggan wuwei JiangXin decoction?fresh/dry/stir-frying?fingerprints were obtained by introducing the data into the Chinese Pharmacopoeia Commission's similarity Evaluation system for chromatographic fingerprints of traditional Chinese Medicine?2012,version?,and the fingerprints were stir-frying.The results of fingerprinting in 10 batches of samples were compared.UPLC-Q-TOF-MS was used to analyze the changes of whole component group before and after processing compatibility in negative ion mode.Combined with Progenesis QI software,principal component analysis was carried out.The chemical constituents of Linggan wuwei JiangXin decoction?fresh/dry/stir-frying?were determined by partial least squares discriminant analysis.3.Effects of fresh,dry and stir-frying combined with Wugan Wuwei Jiangxin Decoction on material energy metabolism in rats with cold drinking pulmonary asthma model and their correlation with spectral effectA rat model of cold-dumping lung type asthma was prepared by using ovalbumin?OVA?sensitization plus ice swimming.Finally,8 normal rats were used as the control group,and the rats with successful modeling were divided into the model group,the positive drug group?Guilong Kechuanning capsule?,the ginger compound group,the dry ginger compound group,and the stir-frying compound group.8 rats in each group.One month after the administration,HE staining was used to observe the changes of lung tissue in rats,and the changes of cAMP?cGMP?cAMP/cGMP?LA?PA?TG?TSH and TRH in each group.Spectral-effect correlation analysis was carried out on the analysis results of mass spectrometry and the investigation index of material energy.Result1.Optimization of decoction of Linggan Wuwei Jiangxin DecoctionThrough the investigation of the content of the index components and the comprehensive weighting method,the water decoction process finally optimized is 12times the amount of water,soaked for 50 minutes,boiled for 40 minutes,and boiled twice.2.Effect of fresh,dry and stir-frying ginger combined with Linggan Wuwei JiangXin decoction on the chemical constituents of the whole prescriptionThrough the traditional Chinese medicine chromatographic fingerprint similarity evaluation software?2012,version?,it is determined that the common fingerprint peaks of raw,dry and stir-frying compound are 23,22,23,respectively,and there are differences in the content of common substances in different compounds.Correspondingly,a total of 44 components were identified by mass spectrometry software,including 35 kinds of ginger compound,35 kinds of dry ginger compound,34kinds of stir-frying ginger compound,and there were differences between different compound components.3.Effects of combined with Linggan Wuwei Jiangxin Decoction?fresh/dry/stir-frying?on material energy metabolism in rats with cold drinking pulmonary asthma model and correlation analysisThe rats in the normal group were sensitive,normal diet and hair luster.The rats in the model group showed irritability,swelling of the limbs,blueness in the lips,shortness of breath and dullness of the hair.The corresponding symptoms of Linggan Wuwei?dry/stir-frying?Jiangxin decoction group were lighter or more normal than those of the model group.Compared with the model group,the content of cAMP?cGMP?cAMP/cGMP?LA?PA?TG?TSH?TRH in Linggan Wuwei Jiangxin decoction?fresh/dry/stir-frying?group had significant difference?P<0.05?.Compared with the corresponding material and energy metabolism index of normal group,there was a significant difference between Linggan Wuwei Jiangxin?frseh?decoction?P<0.05?.Spectral effect correlation analysis was used to identify 18 active components,including3 saponins,4 ginger phenols,5 lignans and 5 flavones.ConclusionTaking the classic prescription,Linggan Wuwei Jiangxin Decoction as the research object,through thechange of chemical composition before and after processing-compatibility of ginger,the energy metabolism of the model animal was affected by the change of chemical composition,and the effects of fresh,dried and processed ginger on the energy metabolism of the model animal were studied.The effects of different processed products on the material basis and efficacy of compound were clarified,and the mechanism of processing-compatibility of ginger was preliminarily elucidated.The research results of the subject will provide reference for the clinical application of ginger artillery products and the mechanism of Linggan Wuwei Jiangxin Decoction in the treatment of cold drink asthma.
