Taking ShenMai decoction, a traditional Chinese medicine, as the object of study, the author of the thesis studied the material basis and mechanism of the compatibility of Ginseng radix and Ophiopogon Japonicas on the basis of UPLC-TOF-MS, which enhanced curative effect and attenuated the effect of the virus. At first, an in vitro study was undertaken to observe Ophiopogon Japonicas’ effect on the digestion of ginsenosides when Ginseng were compatible with Ophiopogon Japonica in different ratios and to expound the material basis for enhancing curative effect and attenuating the effect of the virus when the two medicines were compatible with each other on a chemical level. Based on this study, the method of metabolomics was utilized to make further research in the changes of in vivo endogenous materials as a result of the compatibility of the two medicines. To be specific, healthy SD rats were taken as testees. By means of PCA (principal component analysis) and OPLS-DA (orthogonal partial least squares discriminant analysis), the difference of metabolites in the rats’ urine were analyzed. Then the potential endogenous biomarkers were screened out with VIP (Variable important in projection) and t Test to analyze the relevant pathways of metabolism and to expound the in vivo mechanism of action of ShenMai decoction. Finally, a pathological model was used to further explain the mechanism of ShenMai decoction in treating diseases in clinical practice. To be specific, the method of metabolomics was adopted and the myocardial damaged rats injected with DOX (doxorubicin) were taken as testees. By means of PLS-DA (The partial least squares discriminant analysis) and OPLS-DA (orthogonal partial least squares discriminant analysis), the difference of metabolites in the rats’urine were analyzed. Then the relevant pathways of metabolism were analyzed to expound the mechanism of action of ShenMai decoction in the model of disease.The results of the study showed that when Ginseng was compatible with Ophiopogon Japonica, the amount of dissolution of some ginsenosides of protopanaxadiol type, such as Rb1, Rb3, Rd and their malonyl derivatives was more than the theoretical value; the amount of dissolution of ginsenosides of protopanaxatriol type, such as Rg1, F3 and oleanolic acid ginsenoside Ro was lower than the theoretical value, which indicates that the interaction of Ginseng and Ophiopogon Japonicas changed the amount of dissolution of some ginsenosides in Ginseng after it was compatible with Ophiopogon Japonicas and this would be a probable material basis for the compatibility of Ginseng and Ophiopogon Japonicas to enhance curative effect and attenuate the effect of virus. In the metabolomics study of ShenMai decoction’s effect in the urine of healthy rats, sixteen endogenous metabolites were identified, which included 1-Methyladenine,4,6-Dihydroxyquinoline and p-Cresol glucuronide, which involved the pathways of tryptophan metabolism, histidine metabolism, phenylalanine metabolism, tyrosine metabolism, fatty acides metabolism, purine metabolism and energy metabolomics. These results showed that ShenMai decoction can change physiological functions through some in vivo metabolic pathways so as to be effective. In the study of ShenMai decoction’s function in protecting the myocardial damage caused by DOX, fourteen endogenous metabolites were identified, which includes Succinyladenosine, phenylbutyryglutamine and S-(3-Methylbutanoyl)-dihydrolipoamide-E and indicated that ShenMai decoction protected from myocardial damage by regulating purine metabolism, some amino acid metabolism, fat metabolism and energy metabolism.To sum up, the study was to expound the material basis for the compatibility of the two medicines to enhance curative effect by probing into the compatibility of Ginseng and Ophiopogon Japonica on the one hand. On the other hand, the study explained the mechanism of action of ShenMai decoction in improving the blood supply of heart, liver and other important organs, intensifying microcirculation, enhancing the anoxia tolerance of human body, reducing myocardial oxygen consumption, protecting and repairing myocardial cells, improving the cells’immunity, etc. Thus, the grounds were provided for ShenMai decoction’s reasonable utilization in clinical practice and for treating myocardial damage caused by DOX effectively.
