Objective:Taking drug-couple Psoralea corylifolia L-Myristica fragrants Houtt(PCL-MFH) as research object,from the change of the content to optimize to the best compatibility proportion of drug-couple.Taking multi-indicators as research object and using the orthogonal design to optimization the extraction process.Using UPLC-Q-TOF / MS technology to expand the qualitative analysis of water extraction of the drug drug-couple.Clearing the component information of water extraction.After comparing with the the previous study of alcohol extraction process,clearing the component information of water extraction.Clarify the content differences and the types under the different of solvent extracts and lay the foundation for further development of the drug-couple on the pharmacodynamic studies and the development of new formulationsMethod:1.The research on the change of chemical composition and type in different proportion and types of drug-couple PCL-MFH.The classic compatibility ratio of PCL:MFH is 2:1 and take this ratio as the core,based on consulting literature,design 7 different proportion of drug-couple PCL-MFH,there are 3:1、2:1、1:1、1:0、0:1、1:2、1:3respectively.According to the traditional water extraction technology,extract the main chemical composition.After pre-experiment,we choose the content of psoralen、isopsoralen、isobavachalcone、dehydrodiisoeugenol and bakuchiol as index,seeking the best compatibility proportion of exert the effect.2. Study on the water extraction process of drug-couple PCL-MFH and compare to the alcohol extraction process.To optimaize the water extraction technique for drug-couple PCL-MFHand confirm the optimum extraction process parameters by develop an UPLC fingerprint combined with multi index comprehensive weighted score evaluation.Orthogonal test method was uesd to optimize the process with infiltration time,the amount of water, extraction time,extraction times as detecting factors, the transfer rate investigation of psoralen,psoralen、isopsoralen、 isobavachalcone、dehydrodiisoeugenol and bakuchiol with UPLC fingerprint characteristic peak area as examining indexes,the data were analyzed by multi index comprehensive weighted score evaluation.According to preferred water process conditions to extract the main chemical components of the drug-couple and the total number of peaks and attributionand so on.3. The qualitative analysis of water extraction of drug-couple PCL-MFH and the research of the differences ethanol extracts parts.The qualitative identification of the chemical composition of drug-couple PCL-MFHbased on UPLC-Q-TOF/MS technology.From the tandem mass spectrometry and through the Masslynx V4.1 software,we can obtain the precise molecular,then deduce the possible elements.By the databases of Chemexper and SDBS,then verify the secondary ion mass spectrometry fragmentation of the information matched with the known ingredients,identificate the unknown compounds finaliy.After this part and compared with kinds of total compound species and unknown compounds under ethanol extract.Result:1.Optimize to the best compatibility proportion of drug-couple PCL-MFH is 2:1.2.Optimization to the best water extraction process of drug-couple PCL-MFH:infiltration time 40 min, dosage of water 8 times, each time to extract 1h,extracting 3 times.After comparing to the alcohol extraction process,finding that the three main active ingredient content and the total number peaksbelow the alcohol extraction process andthe dry extract was higher.After the ascribed compared draw the conclution that the differences of the chemical compositioncaused mostly from psoralen,The reseaon may be is that the ingredient of the water-soluble components inincreased or the reseaon may be due to high temperature caused decomposedcomponents,Therefore,when we are extractingthe chemical constituents of the drug pharmacodynamic studies,the chemical properties of the preparation of the index components shuould be considered in order to prefer the extraction process.3.The qualitative identification of the chemical composition of drug-couple PCL-MFHbased on UPLC-Q-TOF/MS technology and identificate 19 kinds of compounds totally,and including 14 kinds of unknown compounds.Afterattributed and compared with the kinds of the total and unknown compounds under the alcohol extraction process.The results show that the common components of the two parts are coumarin,flavonoids,lignans and single quote phenolic.There are still differences in each of compounds of the common parts.The kinds of thecoumarins and flavonoids increased compared with Ethanol extractsites.The other two kinds of compounds just a little change.The coumarin, flavonoid as its main differences ingredient,indicating that the water extraction process and extraction processes have advantages and disadvantages and need further by comparing pharmacodynamic studies.Conclusion:From the change of the content to optimize to the best compatibility proportion of drug-couple PCL-MFH and the result is that psoralen, nutmeg mass ratio of 2:1;After comparing to the alcohol extraction process,finding that the differences of the chemical compositioncaused mostly from psoralen;The qualitative analysis of water extraction of drug-couple PCL-MFH and the research of the differences ethanol extracts parts indicating that although water extraction index component content lower than ethanol extract sites, but its kind of compound is superior to alcohol extraction.Therefore,when we are extractingthe chemical constituents of the drug pharmacodynamic studies,the chemical properties of the preparation of the index components shuould be considered in order to prefer the extraction process.The experiments describes the differences in the water extraction and ethanol extract sitesfrom the perspective of in vitro material basis,to lay the foundation for further research and the development of new formulations...
