Objective:This topic is based on the principle of "eating cold food in cold temperature will hurt the lung",establishment rat model of chronic bronchitis with syndrome of cold fluid retention in lung by injecting lipopolysaccharide,cold stimulation and smoke inhalation.To observe the effect of Linggan Wuwei Jiangxin Decoction on the pathological changes and expression of AQP-1 of lung tissue in rats of chronic bronchitis with syndrome of cold fluid retention in lung,explore the mechanism of Linggan Wuwei Jiangxin Decoction in treating chronic bronchitis with syndrome of cold fluid retention in lung.Methods:1.Forty rats were weighed and randomly divided into four groups:blank group,chronic bronchitis group,traditional Chinese medicine intervention group and cold fluid retention in lung group with 10 rats in each group.2.The rats of blank group were fed normally,the other three groups were injected 3.5%chloral hydrate intraperitoneally on the first and the 14th day,respectively.200 ug(1?g/?l)lipopolysaccharide solution was injected into the trachea quickly,and then the rats were rotated to make the lipopolysaccharide evenly distributed in both lungs.Rats were placed in a smoking box for 2-30 days(except for 14 days).They were smoked twice a day,10 cigarettes were lit for 30 minutes each time,and the interval between the two fumigation was 4 hours.After traditional Chinese medicine intervention group and cold fluid retention in lung group were smoked,cold stimulation was carried out for 12 hours a day until the end of modeling,lasting for 30 days.3.The rats in the blank group and chronic bronchitis group were given 0.9%normal saline by gavage on the 16th day of model establishment.The traditional Chinese medicine intervention group was given Linggan Wuwei Jiangxin Decoction 1g/ml once a day for two weeks.4.After two weeks of administration,fasting water for 12 hours,eyeball blood was taken.Serum levels of IL-2,IL-4 and AQP-1 in lung and bronchus were detected by double antibody sandwich ABC-ELISA method.Histopathology was used to evaluate the inflammation of lung,trachea and bronchus.Results:1.General observation of rats in each group:The spirit of rats in chronic bronchitis group and cold fluid retention in lung group showed different degrees of depression,coughing and sneezing,and sputum sounds could be heard.A little white or yellow viscous secretion appeared in the nose of most rats.2.Histopathological observation:Compared with the blank group,HE staining of lung tissue in other groups showed different degrees of airway injury and inflammatory cell infiltration.In the traditional Chinese medicine group,there was little inflammatory exudation,no proliferation or atrophy of pseudostratified ciliary columnar epithelium of bronchial mucosa at all levels,and occasionally a small amount of mucosal epithelial exudation or erythrocyte exudation of bronchioles and their branches.No edema or hemorrhage was found in the bronchial cavity and alveoli at all levels.3.The results of serum IL-2 and IL-4 of rats in each group:Compared with the blank group,the expression levels of IL-2 and IL-4 in accumulation of cold fluid in the lunggroup and chronic bronchitis group increased significantly(P<0.05);the expression levels of IL-2 and IL-4 in traditional Chinese medicine group increased(p>0.05),but there was no statistical significance;compared with cold fluid retention in lunggroup,the expression levels of IL-2 and IL-4 in traditional Chinese medicine group decreased significantly(P<0.05).4.The results of AQP-1 detection in lung and bronchial tissues of rats in each group:Compared with the blank group,AQP-1 expression in lung and bronchial tissues of rats in cold fluid retention in lunggroup and chronic bronchitis group decreased(P<0.01),while AQP-1 expression in lung and bronchial tissues of rats in traditional Chinese medicine,group decreased(P<0.05),with statistical significance.Conclusion:The model was replicated successfully.The expression levels of IL-2,IL-4 and AQP-1 are correlated with the activity and severity of chronic bronchitis,which can reflect the degree of lung injury and have guiding significance for disease assessment and therapeutic effect evaluation,Linggan Wuwei Jilngxin Decoction can reduce the expression of IL-2,IL-4 and AQP-1,thus regulating the secretion of airway fluid and the expression of inflammauory factors,and play a role in the treatment of chronic bronchitis.This may be one of the mechanisms of Linggan Wuwei Jiangxin Decoction can treat chronic bronchitis with cold fluid retention in lung.
