Background: Idiopathic pulmonary fibrosis(IPF)is the most common idiopathic interstitial pneumonia(IIP),characterized by chronic and progressive fibrosis,irreversible damage to lung function,leading to respiratory failure.Therefore,the diagnosis and treatment of pulmonary fibrosis is very important.Objective: To explore the mechanism of Danbeiyifei Decoction regulating PI3K/Akt/mTOR signaling pathway in the treatment of pulmonary fibrosis and vascular endothelium,so as to provide evidence for clinical treatment of pulmonary fibrosis.Methods: 1.Establish a rat model of pulmonary fibrosis.2.Normal control group(Con group),model group(BLM group),positive drug control group(Pre group),low,medium and high dose Danbeiyifei Decoction group(L,M,H group)were set up.3.The general state,color,volume,shape,elasticity,hardness of lung tissue,morphology,boundary and pathological changes of endothelial cells were observed to determine the degree of alveolar inflammation and pulmonary fibrosis.4.The expression of PI3 K,p-Akt,mTOR protein and mRNA was detected by Western blot and RT-PCR.Result:1.Con group,Pre group and L,M,H group were better than BLM group in mental state,body weight and lung tissue.2.Compared with the Con group,the expression of PI3 K protein and mRNA in BLM group,Pre group and L,M,H group increased significantly on the14 th and 28 th day,and the expression of PI3 K protein and mRNA on the 28 th day was higher than that on the 14 th day(P < 0.05),but the most significant increase was found in the middle dose group of Danbeiyifei Decoction.3.Compared with Con group,PI3 K,p-AKT and mTOR protein and mRNA expression in BLM group and M group increased significantly on the 14 th and28th day,and protein and mRNA expression on the 28 th day was higher than that on the 14 th day(P < 0.05).Conclusion:In the rat model of idiopathic pulmonary fibrosis,Danbe Yifei Decoction up-regulates PI3 K / Akt/mTOR signaling pathway to repair endothelial cells,so as to achieve the purpose of treating idiopathic pulmonary fibrosis,the optimal concentration is 0.6g/ml.
