Study Of Danbeiyifei Prescription On MMP-9 And Egr-1 Expression Efect In Pulmonary Tissue Of Pulmonary Fibrosis Rats

Objective:The purpose of this study is to improve the situation of Dan Beiyifei recipe Decoction on pulmonary fibrosis in rats. By observing the effects of Dan Beiyifei recipe Recipe on pulmonary fibrosis in the lung tissue of rats with MMP-9 and Egr-1 level, Dan Beiyifei recipe on pulmonary fibrosis, and the mechanism is discussed.Method:90 Wistar rats, half male and half female, in accordance with the adaptability of clean grade standard feeding 1 week later, 90 rats according to Literature Zhongbo mycin method modeling, were randomly divided into for six groups, normal control group, model group, Dan Beiyifei recipe low dose group,Tempe to benefit the lung dose group, Dan Beiyifei recipe high dose group,positive medicine control group, 18 rats in each group. Respectively with saline,Dan Beiyifei recipe low dose Decoction(material concentration 0.3g/ml), Tempe to benefit the lung dose water decoction(at different concentrations).Dan Beiyifei recipe high-dose Decoction(concentration 1.2g/ml), prednisone water solution with concentration of 0.5mg/ml), by 10ml/kg body weight filling stomach. After modeling respectively at 7, 14, 28 days were killed 6 rats,drawing, to observe the changes in the appearance of each rat lung,determination of rat lung coefficient, hematoxylin and eosin staining and Masson staining, determine the degree of alveolitis and pulmonary fibrosis,using immunohistochemical method to detect the expression of MMP-9 and Egr-1expression level.Result:Traditional Chinese medicine in each dose group and prednisone group rat lung coefficient comparison between groups, over the same period of model group were lower(P<0.05),the comparison corresponding pathological tissue slice observation,prompt prednisone,Dan Beiyifei recipe height measurement can reduce rat alveolar inflammation degree, reduce fibrosis progression.Prednisone and Chinese medicine dose group rat alveolar inflammation and fibrosis of integral is lower than the same period model group(P<0.05), and further validation of prednisone and Chinese medicine high and low dose of alveolitis and pulmonary fibrosis in rats have therapeutic effect. Compared with the blank group of the same period,the rest of the group of rats lung tissue expression of MMP-9 and Egr-1 were increased(P<0.05),suggesting the formation of pulmonary fibrosis associated with MMP-9 and Egr-1;Prednisone and Chinese medicine dose group from the lung tissue expression of MMP-9 and Egr-1 were lower than that of model group,with statistical difference(P<0.05),Dan Beiyifei recipe party treatment for pulmonary fibrosis mechanism may be related to lower the expression of MMP-9 and Egr-1.General condition of rats during this period,the prednisone group and traditional Chinese medicine each dose group were better than the model group, prednisone and Dan Beiyifei recipe can improve the symptoms of pulmonary fibrosis in rats.Conclusion:Dan Beiyifei recipe can improve the symptoms of pulmonary fibrosis rats,reduce rat lung coefficient, reduce the alveolitis and pulmonary fibrosis degree, reduce the lung tissue of the expression of MMP-9 and Egr-1,and reduce the expression may be Dan Beiyifei recipe one of treatment for pulmonary fibrosis mechanism.
