Effect Of Danbeiyifei Decoction On HSPA12B Expression In Vascular Endothelial Cells Of Rats With Pulmonary Fibrosis

Objective:The effects of Danbeiyifei Decoction on endothelial cell injury markers ET-1 and vWF,and the influence on HSPA12B expression in vascular endothelial cells in rats with pulmonary fibrosis were studied.Furthermore,the protective mechanism of Danbeiyifei Decoction on vascular endothelial injury in rats with pulmonary fibrosis was revealed.Method:108 SPF Wistar male rats were randomly divided into six groups with 18 rats in each group:normal control group(group A),model group(group B),positive drug control group(group C),low-dose group(group D),medium-dose group(group E)and high-dose group(group F).The rats in the normal control group were given 0.9%sodium chloride injection instead of bleomycin to establish pulmonary fibrosis model,while the rats in the other five groups were given 5mg/ml(50%)bleomycin dilution to induce pulmonary fibrosis.The next day after the model was made,rats in the normal control group and the model group were given 0.9%sodium chloride injection(0.2 ml/20g body weight)once a day,and rats in the positive drug control group were given prednisone acetate suspension(0.32mg/ml)once a day.The concentrations of decoction in the low,medium and high dose groups of Danbeiyifei Decoction were 0.3mg/ml,0.6mg/ml,1.2mg/ml,respectively.After giving for four weeks,six rats in each group were taken at two time points of 14 and 28 days after the model establishment,and the rats were killed by cervical dislocation after weighing.Indicators to be measured:ET-1 and vWF in serum were detected by ELISA,HSPA12B was detected by Westen blot and HSPA12B mRNA expression level was detected by RT-PCR.Result:The expression of ET-1,vWF,HSPA12B mRNA and protein in the normal control group was not significantly changed.The levels of ET-1,vWF,HSPA12B mRNA and protein in the model group were significantly higher in each stage than those of the control group(P<0.01).The levels of ET-1,vWF,HSPA12B mRNA and protein in the low,medium and high dose of Danbeiyifei Decoction groups and the positive control group were higher than that of the normal control group(P<0.001),but lower than that of the same period model group.Among them,there was no statistical difference between the low-dose group and the positive drug control group,while the high-dose group had significance compared with the positive drug control group(P<0.01).Conclusion:Danbeiyifei Decoction can inhibit the expression of ET-1 and vWF to protect vascular endothelial function.Danbeiyifei Decoction can protect vascular endothelium by regulating the expression of HSPA12B in vascular endothelial cells of pulmonary fibrosis rats.
