1 PURPOSEDryness is an important function of Atractylodes Atractylodis.This study hopes to establish a more comprehensive method to characterize the dryness of Atractylodis Atractylodis,to confirm the dryness part of Atractylodis Atractylodis,and then to better determine the key components of Atractylodis Atractylodis.Finally,we use the dryness substance of Atractylodis to preliminarily explore the preventive and To provide reference for further expanding the clinical application of Atractylodes macrocephala.2 METHODS 2.1 Study on Dryness Characteristic of Atractylodes macrocephala and Screening of Dry PartsHealthy rats were given high and low doses of Atractylodes rhizoma,bran Atractylodes aqueous extract and volatile oil for 21 days respectively.Based on the theory of dryness,water intake,submandibular gland morphology,urine volume,aquaporin 2(AQP2)expression in kidney and hemorheology were selected as indicators to observe the dryness effect of each group.2.2 Spectrum-effect relationship of volatile oil of Atractylodes macrocephalaGC-MS(gas chromatography-mass spectrometry)was used to analyze the volatile oils of Atractylodes macrocephala(S1-S9)in different batches.The GC-MS fingerprints of the volatile oils were established.The average daily water consumption,whole blood viscosity and AQP2 content in kidney of rats were used as the pharmacodynamic indexes of the dryness effect of Atractylodes macrocephala.Orthogonal partial least squares(OPLS)method was used to analyze the spectral effect relationship.2.3 Pharmacodynamics of Atractylodes Atractylodis Dry Substance on Cerebral Edema Induced by Focal Cerebral Ischemia Reperfusion Injury in MiceC57 mice were divided into four groups: normal group,model group,Atractylodes rhizome volatile oil group and beta-eucalyptol group.Normal group and model group were given saline of equal volume daily,Atractylodes rhizome volatile oil group was given 250 mg/kg of Atractylodes rhizome volatile oil solution daily,beta-eucalyptol group was given 75 mg/kg of beta-eucalyptol solution daily for a continuous period.After 7 days of administration,MCAO model(focal cerebral ischemia model)was established.Neurological function scores,brain water content and cerebral infarction area were measured by TTC staining.Meanwhile,the time and dose dependence of beta-eucalyptol group were studied by three-day and seven-day administration,25 mg/kg and 75 mg/kg dosage.3 RESULTS 3.1 Study on Dryness Characteristic of Atractylodes macrocephala and Screening of Dry PartsCompared with the soybean oil group,the rats in the high dose group of volatile oil of Atractylodes lanceolata had significantly increased water intake(P<0.05),atrophied submandibular glands,increased urine volume(P < 0.05),decreased expression of aquaporin 2(P<0.05),and increased blood viscosity(P<0.05).Compared with the high-dose group of volatile oil of Atractylodes lanceolata,the difference between the high-dose group and the high-dose group was significant.There was no significant difference between the water extract group of Atractylodes macrocephala and bran Atractylodes macrocephala and the saline group.3.2 Study on the spectral-effect relationship of volatile oil of Atractylodes macrocephalaThe dryness effect of Atractylodes lanceolata may be the result of the interaction of various components.The two peaks contributing to the dryness effect are 31 and 33 peaks respectively.The corresponding components of the two peaks identified by spectral library retrieval are beta-eucalyptol and isoguaiacol.3.3 Pharmacodynamics of Atractylodes Atractylodis Dry Substance on Brain Edema Induced by Focal Cerebral Ischemia Reperfusion Injury in MiceAtractylodes Atractylodes volatile oil and beta-eucalyptol have no statistical difference in improving brain edema caused by focal cerebral ischemia brain injury,but beta-eucalyptol has significant improvement on cerebral edema complications and cerebral infarction caused by focal cerebral ischemia brain injury,and has better neuroprotective effect.At the same time,the time-dependent experiment showed that the preventive effect of 3-day administration was not as good as 7-day administration.The dose-dependent experiment showed that both high and low doses had better preventive effect.4 CONCLUSIONIn this study,a comprehensive evaluation system of dryness effect of Atractylodes macrocephala was preliminarily established,which confirmed that the main dryness part of Atractylodes macrocephala was volatile oil.It was revealed that the mechanism of dryness reduction by stir-frying of Atractylodes macrocephala bran was not only related to the decrease of the total content of volatile oil,but also related to the transformation of dryness components in volatile oil.The main dryness components of Atractylodes macrocephala were identified preliminarily,which provided a reference for the basic research on dryness of Atractylodes macrocephala.The preventive and therapeutic effects of Atractylodes dryness substances on related edema were preliminarily discussed,which provided a certain reference for the clinical application of Atractylodes dryness substances.
