The Effects And Mechanisms Of Volatile Oil Of Atractylodes In Acute Liver Injury Of Mice

Liver injury shows abnormal liver function caused by a variety of causes in clinical,and severe and sustained liver damage can lead to liver failure.Atractylodes,an important traditional Chinese medicine,has volatile oil from its roots and stems.The main component of the volatile oil has positive effects on cirrhotic ascites and liver cancer.One of acetanilide,acetaminophen,is a common analgesic antipyretic medicine,while the overused of acetaminophen links to acute liver injury,which in some cases can result in coma and death.In this experiment,after detecting several methods of extraction,we finally decided to use the volatile oil which was got by steam distillation for the following experiments.For the improper using of drugs is a common reason for acute liver injury,acetaminophen which is an ordinary drug to induced acute liver injury in mice.In this study,to detect effects of volatile oil of atractylodes in acute liver injury,we used acetaminophento induced acute liver injury.In addition,mice were given treatment of volatile oil of atractylodes by intragastric administration for a week.Results showed that,in acute liver injury of mice,after treated by volatile oil of atractylodes,the level of serum AST and ALT declined with GSH improved in liver in the acute liver injury of mice.The results of western bolt indicated volatile oil of atractylodes can activate the AMPK and Nrf2 pathway.Moreover,the levels of HO-1and NQO1,which are associated with oxidative stress,were improved.Immunohistochemical confirmed that volatile oil of atractylodes can activate Nrf2pathway.So,we take the same test in Nrf2^-/-mice,the volatile oil of atractylodes protective effects of antioxidant were less compared with wide type mice.Results showed that the volatile oil of atractylotomy could activate the autophagy pathway to protect mice from acute liver injury induced by acetaminophen.Results showed that volatile oil of atractylodes has significant effects in alleviate oxidative stress,which is valuable in detected the mechanism and medicine in liver injury.
