| Tingli Shengmai decoction is composed of Radixginsengrubra,Atractylo-des macrocephala,Ramulus Cinnamomi,Semen Lepidii,etc,which is an experience prescription of Xing Yue-peng treating congestive heart failure(CHF).Atractylodes macrocephala and Ramulus Cinnamomi contain mainly volatile oil of terpenoids and its derivatives.Studies have shown that this kind of composition has many kinds of pharmacological effects,such as strengthening heart,improving myocardial ischemia,inhibiting of myocardial contraction and so on.At present,there is no literature about extraction process.When the volatile oil is extracted together,it is too hard to segregate accurately because of the difference of oil time.Therefore,orthogonal test was used to optimize the extraction process.Nowadays,volatile oil is dissolved directly or added solubilizers to prepare dosage forms.The method is simple and feasible,but the stability is poor.For that reason,the ingredient is liable to mildew and lost.In order to improve the stability of traditional Chinese medicine preparations,the study is to optimize the inclusion technology of volatile oil.Objective: In order to maximize the amount of volatile oil,orthogonal test was used to optimize the technology.In order to ensure the ingredients safety and stability,β-CD and HP-β-CD were used to prepare inclusion compound with mixing method,grinding method,and ultrasonic method.The inclusion complex was verified by TLC and UV.The study can provide the theoretical basis for industrial production of Tingli Shengmai decoction.Methods:1 The volatile oil was extracted with steam distillation.Orthogonal test and comprehensive weight of multi-index to investigated the extraction proces-s.And the index were the amount of volatile oil,atractylode I and cinnamalde-hyde.The amount of water,soaking time and extraction time were the main factors.2 The study was carried out with L9(34)orthogonal design,the inclusion rate of volatile oils,yield of inclusion compound and oil content as the index to evaluate the technologies.The methods of saturated water solusion,grinding and ultrasonic were investigated.The inclusion complex was verified by TLC and UV.Results: 1 The optimum process for extracting enssential oil from Atractylodes macrocephala and Ramulus Cinnamomi was combination of 6 soaking time,1:10solid-liquid ratio,4h extraction time.2 The optimum preparation conditions for inclusion were established as follows: the proportion of volatile oils and β-CD was 1:8,the inclusion time was 3 hours and the inclusion temperature was at 40℃.3 The optimum preparation conditions were established as follows: volatile oil to HP-β-CD =1: 10,the amount of water is 2 times,and the grinding time is 2h.4 The volatile oil inclusion compound was identified buy TLC and UV.The developing solvent was petroleum ether(60℃~90℃)-ethyl acetate(20:1).The chromogenic agent was vanillin-sulfuric acid.UV scanning spectrum of the test solution was in 200 to 400 nm range.The results suggested that the inclusion compound were stable.There was no change about the essential oil of physical and chemical properties.Conclusions:1 The optimum extraction process of water vapor distillation method is reasonable and reliable;It can provide experimental data for the extraction process.2 The inclusion technology is simple and it can improve the stability and solubility of volatile oil effectively.Further more the method can be used for industrial production of Tingli Shengmai decoction. |
