Objective:To investigate the therapeutic effects of gentian total glycosides on Liver Fibrosis in rats.Method: Sixty SD rats were selected and placed in SPF animal laboratory for 7 days for feeding,and then randomly divided into 6 groups(blank group,model group,control group,high,medium and low dose gentian total).All rats were weighed and recorded,except blank group,the remaining groups were injected with 15% CCl4 oil solution intraperitoneally(the dosage of the model was 0.75ml/kg).The dosage of intraperitoneal injection should be doubled at the first time of the model,and the fixed time of the week should be injected three times according to the prescribed dosage.The rats in the administration group were given prescribed doses of drugs immediately after the establishment of the model(the control group was given colchicine according to the standard of 30mg/kg;the high,middle and low dose groups were given total gentian glycosides according to the standard of 100,60,20mg/kg,and were given proper amount of saline after dissolving or dispersing evenly).The rats in the blank group and the model group were given the same volume of physiological salts by gavage.Water.From the 10 th week of the experiment,plasma was centrifuged,serum was separated,total bilirubin and other evaluation indicators were determined;proper amount of liver tissue was extracted,homogenized,supernatant was separated after centrifugation,hydroxyproline and other evaluation indicators were determined;a few left lobe tissues were extracted,pathological sections were made and pathological changes were observed.Result:1.Compared with the model group,the main indexes of liver function in colchicine group,such as liver index,ALT,AST,T-BIL,ALB,GGT,HA,COL III,TGF-beta 1,Hyp,MDA,SOD and inflammatory activity scores,changed significantly(P < 0.01 or P < 0.05),suggesting that colchicine has a certain hepatoprotective effect,possibly by regulating liver index,ALT,AST and other liver function indicators.The main evaluation indexes of liver function can be improved obviously in each dose group,which indicates that gentian has different protective effects on liver.2.Compared with colchicine group,the main evaluation indexes of liver function in high dose group of gentian total glycoside,such as liver index,ALT,AST,T-BIL,ALB,GGT,HA,COL III,TGF-beta 1,Hyp,SOD,MDA and inflammatory activity score,changed significantly(P < 0.01 or P < 0.05),indicating that the dosage of gentian total glycoside was large,the main evaluation index of liver function was improved significantly,and the liver protection effect was better(P< 0.01 or P < 0.05).The changes of hepatic index and GGT in middle dose group and low dose group were significant(P < 0.01 or P < 0.05),while there were no significant changes in other main indexes of liver function(P > 0.05).3.Compared with the high dose group,except for the liver function evaluation indexes COL III,Hyp,SOD,MDA and inflammatory activity score of middle dose group and low dose group,the changes of other liver function evaluation indexes were significant P < 0.01 or P < 0.05).The results showed that the hepatoprotective effect of Gentiana glycosides was related to the dosage.The larger the dosage,the more obvious the improvement of liver function evaluation index and the better the hepatoprotective effect.4.The liver of the model group was enlarged,the volume was larger than that of the blank group,the appearance was relatively yellow,the texture was flexible,the section had a remarkable greasy feeling,the liver tissue necrosis appeared in larger area,the hepatocytes were disorderly arranged,the inflammatory cells infiltrated,and the fibrosis was more than that of the blank group,which indicated that the hepatic fibrosis model had been successfully established;the total gentian glycoside dosage groups could be improved to varying degrees.Liver appearance,reduce liver tissue proliferation,reduce inflammatory cell infiltration,effectively reduce the proliferation of liver fibrosis,the effect and dose have a certain correlation.Conclusion: Gentian total glycosides has a good effect on hepatic fibrosis.
