Objective To evaluate the effect of gentian extractive on the proliferation,apoptosis and expression of the phosphorylation of EGFR(p-EGFR)in the human keratinocyte cell line(HaCaT cells)induced by epidermal growth factor(EGF),and to investigate its possible mechanism in the pathogenesis of psoriasis.Methods Over stimulating HaCaT cells by EGF,then using Inverted Microscope,compare the degree of morphological changes of the HaCaT cells affected by the different concentration gradient of Gentian extracts.Four methyl thiazolyl tetrazolium chromatometry method(MTT)is used to detect the proliferation inhibition of HaCaT cell affected by the medicine.Besides that,flow cytometric is also used to detect apoptosis.In addition,western blot method is used to detect the expression of p-EGFR.Results Gentian extractive inhibited the proliferation of HaCaT cells induced by EGF in a dose-dependent manner,while contrasted with blank control group there has significant differences(P<0.05),in the concentrations of 10-50mg/ml,the proliferation is negative correlated with the concentration(r=-0.991,P<0.01).It promoted the apoptosis of HaCaT cells induced by EGF,positive correlated with the concentration(r=0.996,P<0.05).At the same time,this extractive reduced the phosphorylation of EGFR in HaCaT cells induced by EGF,this effect increased with the concentration.Conclusion One of the mechanisms by which gentian treat psoriasis is inhibiting the over proliferation and promoting the apoptosis of keratinocyte,thus mechanism may achieved by inhibiting the phosphorylation of EGFR so as to blockt the correlative intracellular signal passage.
