Objective:To observe the intervention effect of modified Shengji Yuhong ointment on rat dorsal cortical trauma and explore its intervention mechanism.It was concluded that the modified Shengji Yuhong ointment could promote wound healing by promoting microvascular regeneration and increasing basic fibroblast growth factor(b-fibroblast growth factor).Methods: Using 60 rats were divided into male and female,and the model of skin ulcer was made according to Fu Xiaobing's method of full-thickness skin defect.The models were divided into blank group,Shengji Yuhong ointment group and modified Shengji Yuhong ointment group.All three groups were fed under the same conditions.In the model group,0.9% NS was used to wash the wound for 1 minute,and dry cotton ball was wiped and dried.Gauze dressing.Change dressing once a day.In Shengji Yuhong ointment group,0.9% NS was used to wash the wound for 1minute with uniform low flow rate,and the appropriate thickness of Shengji Yuhong ointment was evenly applied on the wound.Gauze dressing.Change dressing once a day.In the modified Shengji Yuhong ointment group,0.9% NS was used to wash the wound with uniform small flow for 1 minute,and the suitable thickness of the modified Shengji Yuhong ointment was evenly applied on the wound.Gauze dressing.Change dressing once a day.Five animals were randomly selected from each group for execution 3,7 and 14 days after modeling.Immediately after execution,the wound area was recorded by transparent dressing.Then two pieces of tissue 0.5 cm *0.5 cm * 0.3 cm away from the wound margin were fixed in 10% neutral mild formaldehyde for 5 days,and then were embedded in paraffin for reserve.After routine dehydration,embedding and slicing,HE staining was performed to observe the changes of skin and granulation tissue structure under light microscope,and the number of capillaries was observed under 400-fold optical microscope.Immediately after execution,the tissue was immobilized in 10% neutral moderate formaldehydefor 5 days.The tissue specimens were treated with immunohistochemical kit instructions.The number of b-fibroblast positive cells was counted under 400-fold optical microscope.Results:The number of capillaries around wounds in the modified Shengji Yuhong ointment group was more than that in the blank group and Shengji Yuhong ointment group on the 3rd,7th and 14 th days.In the modified Shengji Yuhong ointment group on the 3rd,7th and 14 th days,the number of b-fibroblast positive cells in the surrounding tissues of wounds was more than that in the model group and Shengji Yuhong ointment group.Conclusion:1.Modified Shengji Yuhong Ointment can accelerate the healing of rat dorsal cortical wounds by promoting the proliferation of capillaries around the wound margin,and the effect is due to Shengji Yuhong Ointment.2.Modified Shengji Yuhong Ointment can shorten wound healing time by promoting fibroblast growth and granulation tissue growth.
