| Objective:Observing the effect of Wenshen Gushu Decoction on the expression of Cath K in osteoclasts and Col Ⅰ in bone tissue in ovariectomized rat,to explore the mechanism of Wenshen Gushu Decoction in the treatment of postmenopausal osteoporosis.Method:Establishing the postmenopausal osteoporosis rat model by resection of bilateral ovaries.After successfully modeling,30 SD rats were randomly divided into model group,estradiol valerate group,high-dose of the Wenshen Gushu Decoction group,middle-dose of the Wenshen Gushu Decoction group and low-dose of the Wenshen Gushu Decoction group.Rats in the sham group were excised of adipose tissue around the ovary only.The Wenshen Gushu Decoction high-dose group:[21.21g crude drug/(kg-d)];Middle dose group:[10.605g crude drug/(kg·d)];Low-dose group:[5.3025g crude drug/(kg-d)].Estrogen group:estradiol valerate[10μg/(kg·d)].The model group and the sham group were given saline 0.5ml/100g.Each group was continuously interrupted for 2 months.Then observed bone tissue morphology by HE staining.The content of PICP、ICTP、CTX and NTX in serum were determined by Elisa and the expression of Col Ⅰ and cathepsin K in bone tissue were determined by imunohistochemistry.Result:The experimental results showed that the trabecular bone of the model group was sparse and fractured,which was consistent with the morphological destruction of bone.The trabecular bone structure of the rats in each intervention group was restored,and bone destruction significantly recovered.The efficacy of the Chinese medicine group is related with dose.The efficacy of high-dose group was better than of the middle and low dose groups.Compared with the sham group,serum ICTP,NTX and CTX significantly increased in the model group(P<0.01),but the PICP content was decreased,although it was not statistically significant.Compared with the model group,the serum levels of ICTP,NTX and CTX in the estrogen group were significantly lower(P<0.01),and the PICP content was increased,although it was not statistically significant.The serum levels of ICTP and NTX in the low-dose group of the Wenshen Gushu Decoction decreased(P<0.01)and the content of CTX decreased(P=0.05).The content of PICP increased,but it was not statistically significant.In the high-dose groups and medium-dose groups of the Wenshen Gushu Decoction(P<0.01),the serum levels of ICTP,NTX and CTX significantly decreased and the PICP content significantly increased(P<0.01).Compared with the sham group,the expression of Col I in the bone tissue of the model group decreased(P<0.01)and that of Cath K protein in the bone tissue of the model group significantly increased(P<0.01).Compared to the model group,the expression of Col I protein in bone tissue of the Wenshen Gushu Decoction group and estrogen group increased(P<0.01)and Cath K protein expression of both groups significantly reduced(P<0.01).Conclusion:1.Wenshen Gushu Decoction can effectively improve the osteoporosis state of PMOP model rats in the morphological aspect.2.The treatment of PMOP with Wenshen Gushu Decoction may be achieved by down-regulating the expression of Cath K in osteoclasts and inhibiting the decomposition of Col I in bone tissue. |
