Under the direction of Chinese Medicine Theory and experimental medicine, this study was carried out to investigate the effect of kidney-tonifying formula (KTF) on female ovariectomized rat bone by establishing osteoporosis model and selecting indices through in vivo and in vitro experiments. In vivo experiment, ovariectomized rats were used to establish postmenopausal osteoporosis animal model in which there were 3 testing groups (namely, KTF, Estrogen and Kidney-tonifying single herb) and 3 control groups (namely, age-control, sham and ovx) respectively. The total observation period was 12 weeks. In vitro experiment, KTF drug-containing serum or non drug-containing serum was used to culture osteoblasts from neonatal rat calvaria and the investigation was carried out by cell, molecular and gene level respectively.The experiment results showed that both KTF group and Estrogen group had the same effect on bone, namely increasing the serum calcium level, decreasing the bone turnover. However, Estrogen group was more effective than KTF group in increasing bone mineral density and such results could be related to dosage-dependence and time-dependence. KTF group was better in increasing serum phosphorous level than Estrogen group. KTF group had no effect on the uterus index. KTF drug-containing serum could enhance the proliferation and differentiation of osteoblasts but such effect could be inhibited by Estrogen receptor blocker ICI. KTF drug-containing serum can up-modulate the expression of ER B mRNA in osteoblasts. The experiment indicated KTF's effect on estrogen receptor. Based on the integrated analysis of KTF's effect on bone, for KTF contains no estrogen, such effect could be produced by directly or indirectly affecting estrogen receptor, but can not be produced certainly via estrogen route which needs further discussion and investigation.This study has preliminarily illustrated that KTF could enhance bone formation, inhibit bone resorption and basically, it had the same effect on bone as estrogen. This has preliminarily demonstrated a good promise of KTF on the treatment of osteoporosis.There was discussion in the following 4 parts: (1) the basis of the theory and the creation of the project, (2) the analysis of the results and conclusion, (3) the incomplete exploration and foresight and (4) self-assessment and review. These could give a good reference for future research purpose.
