Objective:The roles of Wnt protein in carcinogenesis have been well documented in human cancers.Wls is a key modulator for the secretion of Wnt protein.This study was aimed to investigate the correlation between Wls protein and the clinicopathological data and prognosis of breast cancer.Methods:In this study four breast cancer cell lines MCF-7,ZR-75,MDA-MB-453 and MDA-MB-468 were tested by qRT-PCR and western blotting(MCF-l0A,a normal human breast epithelial cell,was used as the control).188 breast cancer tissue specimens were subjected to immunohistochemistry.The relationship between the expression of Wls and clinicopathological characteristics and prognosis of patients was analyzed by SPSS.The?~2 test,Spearman,Cox regression,log-rank test and kaplan-meier survival curve analysis were used.P<0.05 was used as the standard for statistically significant difference.Results:By contrast with MCF-l0A,Wls was highly expressed in MDA-MB-453 and MDA-MB-468,and lowly expressed in MCF-7,ZR-75.Wls was lowly expressed in31.4%(59/188),middle in 52.6%(99/188),and highly in 16.0%(30/188)of tested breast cancer specimens.The expression of Wls was positively associated with estrogen receptor(ER)status(p=0.000,r=-0.302),progesterone receptor(PR)status(p=0.000,r=-0.359),Ki-67status(p=0.026,r=0.197)andmolecular classification(p=0.336,r=0.000).COX regression analysis showed that the expression of Wls was positively associated with overall survival(p=0.000).Conclusion:Our data suggest that Wls protein was related to well-known breast cancer markers(ER?PR and Ki67)and OS.When ER(-),PR(-)or Ki67>30%,Wls was highly expressed,and the overall survival time of patients was short.So Wls may be used as a new marker for treatment and prognosis of breast carcinoma.
