Objective:To explore the hypoglycemic effect of water extracts from oyster shell and carambola and its related mechanism so as to provide the experimental basis for delay the occurrence and development of diabetes.Methods:SPF Kunming mice,male,weighing 18-22g.10mice were randomly selected as normal control group.The rest mice were intraperitoneally injected with 120mg·kg-1 streptozotocin?STZ?.After 7 days?12 hours of fasting water before blood collection?,the tail vein blood was collected.The fasting blood glucose?FBG?of mice was measured by Roche's excellent blood glucose meter.The diabetic mice whose FBG(>11.1 mmol·L-1)were regarded as successful models.Diabetic mice were randomly divided into model group,positive control group and muyang high,middle and low dose group with 10mice in each group.On the second day after the success of the model,the rats were given orally once a day for 14 consecutive days.The normal control group and model group were given the same amount of normal saline;the positive control group was grinded and crushed before use,and the suspension was made up of distilled water.The suspension was gavaged with 0.32g·kg-1·d-1;the high,middle and low dose groups of water extracts from oyster shell and carambola were gavaged with 20g·kg-1·d-1,10g·kg-1·d-1 and 5g·kg-1·d-1,respectively.During the administration period,the general conditions of mice,such as mental state,food intake,water intake,stool and urine,hair changes,were observed daily,and the weight changes and deaths of mice were recorded.Serum,thymus,spleen,liver,pancreas and kidney were collected.Fasting blood glucose,fasting insulin concentration,serum total cholesterol?TC?,triglyceride?TG?,high density lipoprotein cholesterol?HDL-C?,low density lipoprotein cholesterol?LDL-C?,superoxide dismutase?SOD?activity and malondialdehyde?MDA?and glutathione?GSH?were measured.HE staining pathological sections of kidney and pancreas were observed.Immunohistochemistry results of Insulin,Bax and Bcl-2 were observed.Result:?1?Before modeling,the mice had dry and smooth fur,bright eyes,good mental state,active action,normal drinking water,food and urine volume,and dry cushion.After modeling,the mice grew slowly,their body was thin,their hair fell off and tangled in clusters to varying degrees,their eyes were dim and dull,their spirits were depressed,their activities were significantly reduced,the consumption of drinking water and feed was significantly increased,their urine and stool were increased,their cushions were wet and smelly,and typical symptoms of"a little more than three"appeared.The drug was administered intragastrically for 14 days.?2?The P value of thymus index in each group was less than 0.05,the difference was statistically significant.There were significant differences between high dose group and low dose group groups;there were significant differences between high dose group,middle dose group,low dose group,positive control,model group and normal control group.?3?The P values of INS and hypoglycemic rate in high dose group,middle dose group,low dose group,positive control group,model group and normal control group were all less than 0.05,with statistical significance.Two indicators were compared.In INS,the P values of the middle dose group,the low dose group,the positive control group and the blank group were all less than 0.05.The difference was statistically significant.It can be concluded that the middle dose group,the low dose group and the positive control group had statistical significance compared with the blank group.?4?The P values of HDL-C,LDL-C,TC and TG were all less than 0.05.The difference was statistically significant.It can be concluded that there were differences in the comparison of HDL-C,LDL-C,TC and TG.There were significant differences between high,middle and low dose groups,positive control group,blank group and model group;LDL-C index found that there were significant differences between low dose group,positive control group,model group,blank group and middle dose group;TG index:middle dose group,positive control group,model group,blank group and low dose group.The difference has statistical significance.There was no significant difference between the other groups.The P values of HDL-C and TG were all higher than0.05,and there was no significant difference.?5?The P values of SOD,MDA and GSH in high,middle and low dose groups,positive control group,model group and blank group were all less than 0.05,with statistical significance.It can be concluded that SOD,MDA and GSH in high,middle,low dose groups,positive control group,model group and blank group were all different.In SOD,the P values of middle dose group,low dose group,positive control group,model group,blank group and high dose group were all less than 0.05.The difference was statistically significant.It can be concluded that high dose of compound peony ointment had an effect on the activity of SOD.In MDA,the P values of low dose group,model group,blank group and high dose group were all less than 0.05.The difference was statistically significant.It can be concluded that high dose of compound peony ointment can increase the content of MDA.In GSH,the P values of middle dose group,positive control group,model group,blank group and high dose group were all less than 0.05,with statistical significance.CONCLUSION:?1?Water extracts from oyster shell and carambola can inhibit the increase of blood glucose in diabetic mice induced by streptozotocin,and improve the body's sensitivity to insulin.The results suggested that the Water extracts from oyster shell and carambola could protect the formation of hyperglycemia and islet injury induced by streptozotocin in mice.?2?Water extracts from oyster shell and carambola can improve SOD activity and GSH content in serum of diabetic mice,and improve spleen index and thymus index in diabetic mice.These results suggested that the Water extracts from oyster shell and carambola could scavenge free radicals,improve the antioxidant ability and enhance the immune function.
