Effect And Mechanism Of The Extracts From Coreopsis Tinctoria On Diabetes Mellitus

Objective:1.To observe the effects of the extracts from C.tinctoria on the lipids and blood glucose in diabetic rats;2.To study the mechanism of the extracts from C.tinctoria in diabetic rats preliminarily.Methods:1.Male SD rats were fed high-fat diet for 6 weeks,then 35mg/kg streptozotocin(STZ)were used to induce diabetic rats by peritoneal injection.The diabetic rats were treated with the alcohol and water extracts about 4 weeks,then FBG?HbAlc?Ins and lipids were determined,the metformin was positive drugs;2.The content of SCr?AST?ALT and 24 h urine mALB were observed,calculate the liver and kidney indexs to evaluate the effect;3.To study the mechanism of the extracts from C.tinctoria in diabetic rats by observe the enzymic activity changes of SOD,GSH-PX and the content of MDA,GLP-1 and adiponectin.Results:1.After 4 weeks,the high and middle doses of both extracts can inhibite the diabetic rats weight loss,reduce the content of Ins?HbAlc?TG?TC and LDL-C(p<0.01)significantly,low the IRI(p<0.01),increase insulin sensitivity,increase the content of HDL-C;2.The high and middle doses of both extracts can low the liver and kidney indexs,reduce SCr?AST and ALT(p<0.01),decrease 24 h urine mALB(p<0.01);3.The high and middle doses of both extracts elevate activity of SOD and GSH-PX(p<0.01),reduce the content of MDA(p<0.01),while also can rise adiponectin,but it dose not affect the content ofGLP-1(p>0.05).Conclusion:1.The extracts from C.tinctoria showed the functions of regulating the lipids,blood glucose level and insulin resistence,and improve the development of renal function abnormal;2.the mechanism of the extracts from C.tinctoria in diabetic rats and renal function abnormal may related with rising the content of adiponectin improveing,anti-oxidative stress and lipid disorders.
