Objective To study the expression of interleukin-24(IL-24)in hepatocelluar carcinoma(HCC)high-incidence family members and carcinoma-free family members,and to explore the correlation between IL-24 and the familial aggregation of hepatocelluar carcinoma(HCC)in GuangXi.Methods 133 members whose families with two or more than two HCC patients were selected as the experimental group and 133 members from families without any cancer were selected as the control group.All of the members were from HCC high incidence areas of GuangXi.It is the matching condition that similar living environment,same gender,same nationality,same HBV carrying statue and age ± 5 years.A total of 266 cases were studied.In the families with high incidence hepatocellular carcinoma there are 67 male members,66 female members,30 Yao nationality members and 103 zhuang nationality members,aged 2 years to 80 years(31.8 ±17.9),52 members younger than or equal to 25 years old,81 members older than 25 years old,25 HBsAg positive members and 108 HBsAg negative members.In the families with carcinoma-free member there are 67 male members,66 female members,30 Yao nationality members and 103 zhuang nationality members,aged 2 yearsto 80 years(31.8 ±17.9),52 members younger than or equal to 25 years old,81 members older than 25 years old,25 HBsAg positive members and 108 HBsAg negative members.In the hepatocelluar carcinoma high incidence families there were 47 cases of first-degree relatives(parents,children,brothers and sisters with the same parents),38 cases of second-degree relatives(grandparents,maternal grandparents,aunts and uncles),and 48 cases of third-degree relatives(Cousins).There were 63 cases coming from families with 2?3 cases of hepatocelluar carcinoma and 70 cases coming from families with 4 or more cases of hepatocelluar carcinoma.Enzyme-linked Immunosorbent Assay(ELISA)was used to detect the expression levels of IL-24 in serum of the experimental group and control group.Results The expression levels of serum IL-24 in family members with high incidence hepatocelluar carcinoma was 304.8751(203.9089,456.2414)ng/L,while that in carcinoma-free family members was 378.3327(268.7921,633.0068)ng/L.There were significant differences between the two groups in the level of IL-24(Z=-3.967,p<0.05).The serum levels IL-24 of male members with high incidence of hepatocelluar carcinoma and those without any cancer were 306.1298(208.4288,404.4768)ng/L and 411.8065(283.3383,609.9527)ng/L,respectively.The differences were statistically significant(Z=-3.629,p= 0.000).The experssion levels of serum IL-24 in female members with high incidence hepatocelluar carcinoma and those without any cancer were299.3691(189.8825,470.9638)ng/L and 372.7160(237.8658,692.7719)ng/L,respectively.The differences were statistically significant(Z=-2.182,p=0.029).The experssion levels of serum IL-24 in zhuang nationality members with high incidence of hepatocelluar carcinoma and those with carcinoma-free families member were 287.2878(208.4288,434.1252)ng/L and 355.5635(245.2594,568.3755)ng/L,respectively.The differences between the two groups were statistically significant(Z=-3.415,p=0.001).The serum levels of IL-24 in yao nationality members with high incidence hepatocelluar carcinoma and those with carcinoma-free family members were 401.1847(188.9516,585.4115)ng/L and 489.6156(384.9068,854.1011)ng/L,respectively.The differences between the two groups were statistically significant(Z=-2.335,p=0.020).The serum levels IL-24 in members with high incidence of hepatocelluar carcinoma and those with carcinoma-free family members aged less than or equal to 25 years old were 301.4240(181.2504,452.2136)ng/L and397.0964(274.5626,646.3215)ng/L,respectively.The differences between the two groups were statistically significant(Z=-2.504,p=0.012).The serum IL-24 levels in members with high incidence hepatocelluar carcinoma and those with carcinoma-free family members aged over 25 years old were 304.8751(220.0605,458.0635)ng/L and 378.1033(265.2100,605.5604)ng/L,respectively.The differences between the two groups were statistically significant(Z=-3.063,p= 0.002).The experssion levels of serum IL-24 in positive HBsAg members with high incidence of hepatocelluar carcinoma and those with carcinoma-free families member with positive HBsAg were 184.2202(151.0795,421.4795)ng/L and 360.5936(190.4188,676.0860)ng/L,respectively.The differences between the two groups were statistically significant(Z=-3.108,p=0.002).The experssion levels of serum IL-24 in negative HBsAg members with high incidence of hepatocelluar carcinoma and those with carcinoma-free families member with negative HBsAg were 315.1265(240.8756,462.7725)ng/L and 385.6720(281.0579,609.5790)ng/L,respectively.The differences between the two groups were statistically significant(Z=-3.047,p=0.002).Irregardless of gender,age,nationality and HBsAg carryingstatus,there were still significant differences between the members with high incidence hepatocelluar carcinoma and the members with carcinoma-free families.In the high incidence hepatocelluar carcinoma families,the serum IL-24 levels of male and female were 306.1298(208.4288,404.4768)ng/L and299.3691(189.8825,470.9638)ng/L respectively,there was no statistical difference between the two groups(Z=-0.115,p>0.05).The serum levels of IL-24 at ages less than or equal to 25 years old were 301.4240(181.2504,452.2136)ng/L.And the serum levels of IL-24 at ages more than 25 years old were 304.8751(220.0605,458.0635)ng/L.There was no statistical difference between the two groups(Z=-0.284,p>0.05).The serum levels of IL-24 of yao nationality and zhuang nationality were 287.2878(208.4288,434.1252)ng/L and 401.1847(188.9516,585.4115)ng/L respectively,with statistically significant differences between the two groups(Z=-2.048,p<0.05).The serum levels of IL-24 of HBsAg positive and HBsAg negative groups were 184.2202(151.0795,421.4795)ng/L and 315.1265(240.8756,462.7725)ng/L respectively.The difference between the two groups was statistically significant(Z=-2.519,p<0.05).In carcinoma-free members from HCC high-incidence families,the serum levels of IL-24 of the first-degree relatives,second-degree relatives and third-degree relatives were 252.0965(161.3105,450.2621)ng/L,320.6206(251.4095,548.9994)ng/L and 309.2690(240.8756,463.8382)ng/L respectively.The results of statistical analysis showed that the serum levels of IL-24 of the first-degree relative was significantly lower than that of the second-degree relative(Z=-2.276,p<0.05),the serum levels of IL-24 of the first-degree relative was significantly lower than that of the third-degree relative(Z=-2.166,p<0.05),and there was no significant difference between the serum levels of IL-24 of the second-degree relative and that of thethird-degree relatives(Z=-0.174,p>0.05).The serum levels of IL-24 in the families with 2?3 cases of hepatocelluar carcinoma and the families with 4 or more cases of hepatocelluar carcinoma were 404.4768(184.2202,605.1554)ng/L and 277.1630(206.1688,338.0225)ng/L respectively.The expression levels of serum IL-24 in family members with 2 to 3 cases of hepatocelluar carcinoma was significantly higher than that of family members with 4 or more cases of hepatocelluar carcinoma,and the difference was statistically significant(Z=-3.003,p=0.003).The levels of IL-24 declined with the increase in the number of HCC patients in HCC high incidence families(p<0.05).According to Multiple Linear Regression(mLR),in carcinoma-free members from HCC high-incidence families,the factor which affected the serum levels of IL-24 were nationality(t=-2.689,p=0.008),HBsAg carrying status(t=3.012,p=0.003),the degree of relation(t=-3.545,p=0.001),the number of HCC patients in the families(t=3.004,p=0.003).Conclusion1.The expression levels of IL-24 may be correlated with the familial aggregation of hepatocellular carcinoma in guangxi.2.The HBV infection and IL-24 may also be related to the familial aggregation of hepatocellular carcinoma in guangxi.3.The expression levels of IL-24 was affected by family history with hepatocellular carcinoma and relative degree,suggesting that genetic factors play a role in the familial aggregation of hepatocellular carcinoma in guangxi.
