Objective:Lung cancer(LC)is the most common malignancy and one of the leading causes of cancer death in the world.More than 80%patients with lung cancer are non-small-cell lung cancer patients.This study aimed to explored the associations between the four single nucleotide polymorphisms in the CD25 gene and NKG2D gene and the occurrence and development of non-small cell lung cancer.Methods:In this study,we recruited,467 patients who were diagnosed with non-small cell lung cancer as case group and 384 healthy people as control group from the first affiliated hospital of kunming medical university and the third affiliated hospital of kunming medical university.Combining with the 1000 Genomes Browser,four single nucleotide polymorphisms(SNPs)in CD25 genes and NKG2D genes were selected with TaqMan probe genotyping.The SNPs loci are:CD25 SNP-rs7072793(C>T),CD25 SNP-rs3118470(T>C),NKG2D SNP-rs2255336(T>C),NKG2D SNP-rs2617160(T>A)?And then the frequency distribution difference of the allele,genotype and haploid type in the case group and the control group,stage ? +II group and ? + ? group,stage ? + ? group and the control group,stage?+? group and the control group were investigated.Results:(1)The results showed that there was no significant difference of allelic frequencies in the 2 polymorphic loci rs7072793 and rs3118470 between all the groups(P>0.05);After the genetic model analysis of the above two loci,it was found that after optimal model Recessive analysis on rs7072793 loci,there were statistically significant differences of frequency distribution in genotype CC-CT and TT between pathological stage ? + ? group and pathological stage III+IV group(CC-CT vs TT,OR = 0.45,95%CI:0.25?0.80,P = 0.004).After optimal model dominent analysis on rs3118470 loci,there was statistically significant differences of frequency distribution in genotype CC-CT and TT between pathological stage ?+ ? group and pathological stage ?+? group(CT,CT vs TT,OR = 1.79,95%Cl:1.09?2.95,P ?0.019).There was no significant difference of haploid type frequencies in the 2 polymorphic loci rs7072793 and rs3118470 in the gene of CD25 between all the groups.(P>0.05).(2)There was no significant difference of allelic frequencies in the 2 polymorphic loci rs2255336 and rs2617160 in the gene of NKG2D between all the groups.(P>0.05);After optimal model Recessive analysis on rs2255336 loci,there was statistically significant differences of frequency distribution in genotype CC-CT and TT between pathological stage ? + ? group and pathological stage ?+? group(CC-CT vs TT,OR = 0.26,95%CI:0.06?1.16,P = 0.040);After optimal model Recessive analysis on rs2617160 loci,there was statistically significant differences of frequency distribution in genotype CC-CT and TT between pathological stage ? + ?group and pathological stage ?+? group(TT-AT vs A A,OR = 0.60,95%CI:0.36?0.99,P = 0.041);There was no significant difference of haploid type frequencies in the 2 polymorphic loci rs2255336 and rs2617160 in the gene of NKG2D between all the groups.(P>0.05).Conclusion:The results of this study indicate that rs7072793 and rs3118470 in the gene of CD25 and rs2255336 and rs2617160 in the gene of NKG2D may be related to the development of lung cancer.In CD25 gene,the polymorphic loci rs7072793 genotype CC-CT may be a protective factor of lung cancer development,but the polymorphic loci rs3118470 genotype TT may be a risk factor of lung cancer development.In NKG2D gene,the polymorphic loci rs2255336 genotype CC-CT may be a protective factor of lung cancer development,and the polymorphic loci rs2617160 may be a protective factor of lung cancer development too.
