Daphne konreana Nakai,also known as Lagencao,the crude drugs is named "Zu Shima",belongs to small shurbs of Daphne.Whole herb and its rootstocks are used to treat diseases,its nature is warm and flavor is pungent.The key function is by-pass network with a warm and stopping blood and pain.Its stem bark is used for the treatment of injuries from falls and bruises in folk medicine.Clinic for mainly treat coronary heart disease,rheumatism,thromboangiities obliterans and so on.Source identification,character identification,microscopic identification,as well as physical and chemical identification methods were performed Systematically on Pharmacognostical study of D.koreana.With Daphnetin and Daphnoretin as reference,to establish qualitative identification by thin layer plat of silica gel GF254.The column was Promosil C18(4.6 mm×250 mm,5 ?m).The mobile phase acetonitrile(B)-0.05%phosphoric acid(A)solution,gradient elution to establish HPLC fingerprint.Flow rate was 1 mL/min.The wavelength was set at 327 nm and column temperature was 40?.The HPLC chromatograms of 10 batches of D.koreana from different habitats were analyzed with similarity calculation by the "traditional Chinese medicine fingerprint similarity calculation software of 2004",the chemometric method was used to assess the quality of D.koreana.In this paper,the content of the extract of D.koreana samples was determined by HPLC,with Daphnetin,Umbelliferone and Daphnoretin.Results:Detailed descriptions for medicinal materials of D.koreana were given including root,stem,leaf transverse section,powder characteristics.The TLC fingerprint of the D.koreana was preliminarily established,TLC spots were clear and well separated and two common spots were better reproducible at 254 nm.The datum of 10 batches of D.koreana samples obtained from different places was analyzed by Chinese medicine chromatographic fingerprint similarity evaluation system 2004A version,and calibrated with 34 characteristic peaks,established a common pattern of HPLC fingerprint,and 3 peaks of them were identified using standard references.The similarity value of these 10 batches of drugs was all above 0.940,indicating a relatively stable quality.The samples were classified into two groups by hierarchical cluster analysis combined with principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA),and fourteen components were the main markers that cause differences in the different batches of samples.Determination of HPLC-UV content:all of the 10 batches of D.koreana herbs contained the three compounds respectively Daphnetin,Umbelliferone and Daphnoretin,the contents were between 21.52?27.19 mg/g,0.45?0.93 mg/g and 5.84?13.20 mg/g respectively.In this study,source identification,character identification,microscopic identification were performed.The TLC and HPLC fingerprints methods were simple,accurate,applicable,and with good stability and good repeatablility.The content determination data was true and reliable.The experimental results mentioned above could be used as the quality standard of the D.koreana.
