Twenty-Five samples of Pinellia which is one of the commonly used traditional Chinese medicines (TCM) were collected. The chemical data was obtained by RP-HPLC, using 0.2% triethylamine(adjusted pH to 3.5 by phosphoric acid )-acetontrile(50:1) and 0.2%triethylamine(adjusted pH to 3.5 by phosphoric acid) as the mobile phase, and detected at 21 Onm. Thirty-three chemical variables were obtained and studied by several chemical pattern recognition methods. All samples were identified with hierarchical clustering analysis and divided into two categories, crude pinellia and pinellia prepratae. The quality of crude pinellia samples was divided into three grades, the superior, the general and the inferior. The correct rate of species indentification and quality grading assessment tested by step discriminant analysis was 100%. A quality grading assessment scheme of Pinellia samples by HPLC was proposed and the results were satisfying.A reversed-phase HPLC method was established to determine the content of trigonelline in 25 Pinellia samples. The mobile phase was 0.2% triethylamine(adjusted pH to 3.5 by phosphoric acid) and detection wavelength was set at 264nm. The results showed the calibration curve was linear in the range of I.86棐37.20 j.tg Emil and correlation coefficient was 0.9998. The average recovery was 96.4%. RSD was 2.1%. This method was simple, sensitive and feasible, and provided a quantitative basis for the quality assessment of RHIZOMA PINELLIA.Relieving cough action was expedited by Pinellia samples, the results showed pinellia has evidental relieving cough action.And relieving3ABSTRACTcough action was expedited by diffierent elution of water deeocta, the results showed alkaloide in chloroform layer and water layer has evidental relieving cough action. Actute toxicity action of pinellia was expedited, the results showed LD50 was 1.33 ?O.19g/kg by injected cavity supension, and water decocta had no toxicity.A new methodology for the quality control of Pinellia samples has been proposed in this study. The method should be helpful for the quality assessment of the TCM.
