24 samples of Radix Bupleuri were collected from different habitat and the quality assessment was developed.(1) Two reversed phase HPLC methods were developed for determination of Saikosaponin a, rutin and quercetin in Radix Bupleuri from different habitats, respectively. In the method for the determination of Saikosaponin a, the Diamonsil C18 column (200 mm × 4.6 mm i.d., 5μm) was used with the mobile phase being methanol-water-THF (70:30:1.5, v/v), flow rate was 1.0 mL·min-1, detecting wavelength being 208 ran, the standard curve of Saikosaponin a was linear in the range of 0.0408~0.408 mg·mL-1 (r=0.9998),The average recovery was 99.7% with RSD 2.0%; An HPLC method was developed for the determination of rutin and quercetin in Radix Bupleuri, the Diamonsil C18 column (200 mm × 4.6 mm i.d., 5 μm)was used with the mobile phase being methanol-water-phosphoricacid (41:59:0.05, v/v), flow rate was 1.0 mL·min-1, detecting wavelength being 370nm,The line arranges were 0.0610~976mg·mL-1 (r=0.9999), 0.0082~0.164mg·mL-1 (r=0.9996),respectively.
