Flos Lonicerae, a famous Traditional Chinese Medicines (TCM), is the dried flower of Lonicera japonica Thunb. , Lonicera hypoglauca Miq. , Lonicera confusa DC. and Lonicera dasystyla Rehd.. It has the effects of bacteriostasis, anti-virus, anti-inflammation, antioxidant and hematischesis. In this thesis the quality control of Flos Lonicerae and the pharmacokinetics of chlorogenic acid in rabbits were studied.The chemical variables as a whole were obtained by HPLC fingerprint analysis and 15 common peaks were detected. The 20 samples were divided into 7 categories by using Hierarchical clustering analysis. Using the data of herbs in I category as standard, Similarity evaluations of all herbs were carried out and the results showed that high-quality herbs' similarities in Lonicera japonica Thunb. were greater than 0.9, the common herbs varied from 0.7 to 0.9, other species' similarities were 0.7 and 0.8 or so. A test of bacteriostasis was carried out and the results showed that the bacteriostasis activity of L. japonica Thunb. was stronger than others. The correlation between the variables and the in vitro bacteriostasis activity was analyzed by regression. Among the chemical variables in the HPLC chromatograms, seven were positively correlated with the bacteriostasis activity.By means of TLC, chlorogenic acid in L. japonica Thunb. and compound Da QingYe injection was identified. A reversed-phase HPLC method for the determination of chlorogenic acid in Flos Lonicerae was used.The samples were separated on an BDS column with a mobile phase of acetonitrile-0.2% H3PO4 (10:90) adjusted to pH3.0 with sodium hydroxide. The flow rate was 0.8ml/min, and the UV detector was set at 327nm. Samples from 20 sources were measured and the contents of chlorogenic acid were within 1.56-5.67%, all met the criterion of the Pharmacopoeia of People's Republic of China, but there are 3 species which are out of the Pharmacopoeia of the People's Republic of China, we could not classify and assess the quality of Flos Lonicerae only according to the contents of chlorogenic acid. A HPLC fingerprint method was developed in our study, the chemical data as a whole were analyzed with Hierarchical clustering analysis and Similarity analysis, through which we could classify all herbs and assess the quality of Lonicerajaponica Thunb.Chlorogenic acid, a major active constituent of Flos Lonicerae was determined in rabbit plasma by HPLC. The plasma concentration-time profile of chlorogenic acid after single i.v. the compound to rabbits was most appropriately described by a two-compartment model, the distribution and metabolism were rapid with half-life of 1.73 l.5min, 12.8 1.1min respectively. Pharmacokinetic data in rabbit after an oral administration of Flos Lonicerae extract were analyzed based on the Statistical Moment Theory., The absorption of chlorogenic acid was rapid with a Cmax of 0.839 0.35 g/ml at 34.7 l.lmin, and a second peak in the plasma concentration of chlorogenic acid of 0.367 g/ml was observed at 274 40min. This phenomenon may be relevant to hepatoenteral circulation. Further investigation is needed to clarify this pharmacokinetic behaviour of chlorogenic acid from the administration of Flos Lonicerae extract.
