Objective:To observe the therapeutic effect evaluation of spleen deficiency in patients withT2 DM and the effect of spleen-heating method on the expression of mTORC1 in rat pancreatic tissue.Methods: Thirty patients with T2 DM were classified as “Spleen Deficiency” and were randomly divided into experimental group(Jianpi Xiaoke Recipe+Amory)and control group(Amory).The main efficacy was: blood glucose(fasting plasma glucose(FPG),2h postprandial blood glucose(2hPG),glycated hemoglobin(HbA1c)),islet function(fasting insulin level(INS),insulin resistance index(HOMA-IR),insulin secretion index(HOMA-?)).Secondary efficacy: blood lipids,body mass index and TCM syndrome scores were used to evaluate the effects of relevant indicators before and after treatment.Animal studies: Wistar male rats were fed with high-sugar high-fat diet and intraperitoneal injection of streptozotocin.After establishing a rat model of T2 DM,the effects of Jianpi Xiaoke Recipe on blood glucose,blood lipids,body weight,insulin function(fasting insulin,insulin resistance index,and insulin secretion index)and other indicators in type 2 diabetic rats were evaluated.Morphological changes of rat islet cytology,fluorescence PCR to observe the expression of mTORC1 in pancreatic tissue.Results: Jianpixiaoke can improve blood glucose,blood lipids,HbA1 C,body mass index,and increase insulin sensitivity and improve islet function in T2 DM patients with spleen deficiency and fever;Expression of mTORC1 in the pancreas serves to protect islet ? cells.Conclusion: Through clinical observation and animal experiments,Jianpixiaoke can improve islet function in patients with type 2 diabetes(Spleen deficiency fever)and has better safety.The mechanism may be to activate the expression of mTORC1 in the pancreas and to protect the islet ? cells. |
