The Difference Expression Of Microrna Of Chronic Hepatitis B With The Syndrome Of Damp-heatin And Deficiency Of Spleen And Damp-heatin Of Spleen-stomach

Objective Study the relationship between the syndrome of damp-heatin and deficiency of spleen and damp-heatin of spleen-stomach of CHB and the differential expression of micro RNA.The purpose is to validate the preliminary screening results of micro RNA gene chip,finding the special micro RNA of two syndrome types of CHB and understanding the regulation mechanism of the special micro RNA,then providing objective biological basis for syndrome differentiation of CHB,aiming at the construction of standardized and objective standard of the CHB syndrome.Methods Extract from the result of our previous studies of the syndrome of damp-heatin and deficiency of spleen of CHB 4 micro RNA(hsa-mi R-1587,the hsa-mi R-4800-5p,hsa-mi R-4466,hsa-mi R-494).And screened out 2 micro RNA(mi R-1273g-3p ?mi R-4419b)with statistically significant different expression in the damp-heatin of spleen-stomach of CHB.Used Real-time fluorescence quantitative PCR Taq Man probe technology,I validated the differential expression of the 6 mi RNAs in 26 cases in patients with CHB(including 13 cases with the syndrome of damp-heatin and deficiency of spleen,13 cases with syndrome of dampness-heat of spleen-stomach)and 13 healthy people.SPSS 19.0 software was used to analyze the measurement data.Results1.Compared with control group,the expression of hsa-mi R-494?hsa-mi R-4466 of the syndrome of damp-heatin and deficiency of spleen group were over-active.There were obvious differences between those two groups(P<0.05);While the expression of hsa-mi R-1587 ? hsa-mi R-4800-5p were down regulation.There was unnoticeable differences between those two groups(P>0.05).2.Compared with control group,the expression of mi R-1273g-3p in syndrome of dampness-heat of spleen-stomach was up regulation.There was obvious differences between those two groups(P<0.05);And the expression of mi R-4419 b was also up regulation,but the difference was unnoticeable(P>0.05).Conclusions1.This study found there were two micro RNA in chronic hepatitis B patients with the syndrome of damp-heatin and deficiency of spleen:hsa-mi R-494,hsa-mi R-4466.The results of this experiment were consistent with previous studies,and the expression of these two micro RNA related to chronic hepatitis B the syndrome of damp-heatin and deficiency of spleen was regulated by the involvement of immunoregulation,inflammatory reaction,P38 lightning MAPK signaling pathways and pd-1 / pd-L1 pathway.Therefore,it can be inferred that the above two micro RNAs could be the diagnostic basis for the morphology and molecularbiology of spleen deficiency dampness and heat syndrome of chronic hepatitis B.2.This study showed that there existed 1 special mico RNA in the plasma of CHB patients with syndrome of the dampness-heat of spleen-stomach : mi R-1273g-3p,the result was consistent with the micro RNA gene chip screening,possibly through to participate in such as the immune response,inflammatory injury,protein metabolism process.Therefore,it can be inferred that mi R-1273g-3p could be regarded as diagnostic basis for the morphology and molecularbiology of dampness-heat of spleen-stomach of chronic hepatitis B.
