ObjectiveValidation specific serum proteomics on Chronic hepatitis B with Liver-stagnation and Spleen-deficiency (GYPX) and Spleen-stomach dampness-heat Syndrome (PWSR) that team early using iTRAQ combined LC-MS/MS technology screening. First to get the history of the GYPX and PWSR and their nature of modern research, as well as the problems and shortcomings, to provide theoretical reference for the research of experiment.Secondly, proposed using Western Blot technique to screen out FETUA, PLTP and PF4V et.al five specific proteins expression for validation, to verify its reliability.MethodsThis article mainly from two aspects of literature analysis and experimental research.First of all, to change the two certificate of relevant literature, spleen and stomach, hot and humid for vertical and horizontal research.For longitudinal study were to explore two certificates of origin context and history;Cross-sectional study:adopt the method of combining the title and abstract in tsinghua tongfang series of CNKI database retrieval two nature of justification of the research literature,retrieve any date for the start date to January 1,2016.157 into GYPX, PWSR 129,and incorporated into the literature from the annual distribution, related diseases, such as molecular studies for measurement and frequency analysis. The protein expression spectra and literature analysis the results and other omics research results summary analysis found that some of the pathways and protein function has the similarity, but did not coincide.CHB change the molecular mechanism of GYPX and enzyme activity regulation, immune response,chemokines mediated signal transduction pathways, platelet activation functions;The molecular mechanism of CHB PWSR with inflammation, lipids, immune response, and other functions.Conclusions1. Modern two certificates certificate to the nature of the results of the study is relatively abundant, but also has the problem of larger diversity and specificity is poorer, so should try to establish the combined disease and liver spleen deficiency, spleen and stomach, hot and humid card multi-index joint diagnosis;In addition,with the integrity of omics technology research characteristic unfolds, it is widely used in the future study of TCM syndrome essence is the trend of The Times.2. After a preliminary screening and the validation, PF4V, PSMA6 as CHB liver spleen deficiency syndromes specific protein expression is more reliable;FETUA, CATC, PLTP as CHB taste hot and damp card syndrome specific protein expression is more reliable.3. For CHB GYPX and PWSR and other profiles of protein expression and our other groups exist some relationship between learning achievement, but has not fully overlap, so the next step should be used to justify syndrome curative effect drug intervention, to dynamically observe the index of the change.
