ObjectiveUseing iTRAQ method to study on different expression correlation between liver stagnation and spleen deficiency syndrome and spleen and stomach damp-heat syndrome of chronic hepatitis B to find the different proteins in each group, to explore the possible biological markers in the two syndromes,then provide a objective basis of differentiation for chronic hepatitis B to promote the development of this disease in TCM.MethodsApplying iTRAQ technology to test the chronic hepatitis B patients with syndrome of liver stagnation and spleen deficiency and spleen and stomach damp-heat,to observe the differential expression between the two syndromes.All dates are analyzed with the SPSS 19.0 software, meanwhile using Mascot system to select the proteins and functional analysis.Results1.Compared with the control group,157 differently expressed proteins were found in liver stagnation and spleen deficiency group (P<0.05),112 proteins were up regulation,45 proteins were down regulation; 163 differently expressed proteins were found in liver spleen and stomach damp-heat group (P<0.05),128 proteins were up regulation,35 proteins were down regulation.2.1n comparision of the two Syndrome groups,30 differently expressed proteins were found in liver stagnation and spleen deficiency group (P<0.05),22 proteins were up regulation,8 proteins were down regulation. Found 1 protein in more than two times:TUBA1A was up regulation;24 differently expressed proteins were found in liver spleen and stomach damp-heat group (P<0.05),22 proteins were up regulation,2 proteins were down regulation. Found 6 proteins in more than two times: ACTA2、ACG2、 ZNF628、DEFA1、DEFA3、CaM were all up regulation.3.Compared with the functions between different proteins and microRNAs, enzyme regulator activity、metabolic process、cellular process、apoptosis、 inflammation response、immune system process may have contacts with the syndrome of liver stagnation and spleen deficiency;immune system process、cell killing、 apoptosis、 response to stimulus、actin may have contacts with the syndrome of liver spleen and stomach damp-heat.Conclusions1.There exist different proteins between syndrome groups and the control group.2. TUBA1A may be the biomarker in chronic hepatitis B with syndrome of liver stagnation and spleen deficiency;ACTA2、ACTG2、ZNF628、DEFA1、DEFA3、 CaM may be the biomarkers in chronic hepatitis B with syndrome of liver spleen and stomach damp-heat.3.In the two syndrome groups, there have same functions between different proteins and microRNAs.4.ITRAQ may possible provide the reference for the future research of chronic hepatitis B. |
