ObjectiveTo further validate the microRNA contained significant differential expression in pre-screening results by the qRT-PCR(fluorescence real-time quantitative method)and compare the changes in the expression of the target microRNA with the pre-screening results.Finally,obtain the characteristic genes of chronic hepatitis B spleen-stomach damp-heat syndrome and chronic-gastritis spleen-stomach damp-heat syndrome,further clarify the biological basis and scientific connotation of spleen-stomach damp-heat syndrome from the perspective of genetics,and explore the potential biomarkers of “the different diseases with same syndrome” in traditional Chinese medicine,in order to provide a scientific and objective basis for the diagnosis of TCM syndromes.MethodsChoose four significant differential expressed genes selected in pre-screening results,the 4 microRNAs co-expressed in chronic hepatitis B spleen-stomach damp-heat syndrome and chronic gastritis spleen-stomach dampness-heat syndrome:hsa-miR-483-3p,hsa-miR-223-3p,hsa-miR-4419 b,hsa-miR-1273g-3p.The subjects were chronic hepatitis b,chronic gastritis,chronic gastritis,spleen and stomach dampness and heat syndrome group and healthy control group,13 cases each.And use the qRT-PCR(fluorescence real-time quantitative method)to analyze the microRNA differential genes detection in blood samples from patients with chronic hepatitis B and chronic inflammatory spleen-stomach damp-heat syndrome.All data in this study are statistically analyzed by using SPSS 20.0 software.Results1.Compared with the healthy control group,the expression of hsa-miR-483-3p and hsa-miR-1273g-3p in chronic hepatitis B spleen-stomach Damp-Heat syndrome is up-regulated.The differences are statistically significant(P<0.05),which are consistent with the pre-screening results;hsa-miR-223-3p,in chronic hepatitis B spleen-stomach Damp-Heat syndrome is up-regulated.The differences are statistically significant(P<0.05),which is not consistent with the pre-screening results;hsa-miR-4419 b has no statistically significant difference(P>0.05).2.Compared with the healthy control group,the expression of hsa-miR-483-3p in chronic gastritis spleen-stomach Damp-Heat is up-regulated.The differences are statistically significant(P<0.05),which are consistent with the pre-screening results;hsa-miR-223-3p、hsa-miR-1273g-3p、hsa-miR-4419 b have no statistically significant difference(P>0.05).3.There was no significant difference in hsa-miR-483-3p,hsa-miR-223-3p,hsa-miR-1273g-3p and hsa-miR-4419 b between chronic spleen-stomach damp-heat syndrome group and chronic gastritis spleen-stomach damp-heat syndrome group(P>0.05).Conclusions1.This study verifies that hsa-miR-483-3p has a certain correlation with the spleen-stomach dampness-heat syndrome and may be an objective diagnostic marker for spleen-stomach damp-heat syndrome.2.Hsa-miR-1273g-3p has some correlation with chronic spleen-stomach dampness-heat syndrome of chronic hepatitis B,and may be biological markers of spleen-stomach dampness-heat syndrome of chronic hepatitis B.3.This study obtained the spleen-stomach dampness-heat syndrome,confirmed that "the same syndrome of different diseases" does exist between the same or similar mechanisms,from the perspective of mirnas further proven biological basis and scientific connotation of taste hot and damp card.
