Shufeng Jiedu Capsule?SFJD?is a traditional Chinese medicine developed by the well-known traditional Chinese medicine practitioner Chu Xian.It consists of Polygonum cuspidatum,Forsythia suspense,Dahuria patrinia herba,Bupleuri radix,Verbenae herba,Isatidis radix,and Phragmitis rhizome,Glycyrrhiza.It has the functions of disparsing wind and clearing heat,removing toxin and easing the throat.It is an important Chinese herbal medicine for the treatment of acute upper respiratory tract infections.At present,researches on SFJD were mostly found in clinical applications,quality control studies of fingerprints,however,there were few reports on the basic research of its effective components,and no report had been made on the basis of the effective components associated with the"drug property and five tastes"of Chinese medicine.Aims:Based on the trilism of“property-response-component”,the material basis of the SFJD was explained by the“spectral-effect”method.Method:1.A uniform design method was used to obtain 22 different matching groups of 8 herbs in SFJD.On the basis of the original capsule preparation process,22 samples of lyophilized powder were obtained.The LC-MS spectral-library of 22samples was established using UPLC-MS technology.2.SFJD has the effect of disparsing wind and clearing heat,and this function mostly spreads heat on the skin surface by promoting sweating and venting.M3 acetylcholine receptors are mainly distributed in the glands,which control glandular secretion and are closely related to sweating.Therefore,Chinese hamster ovary cancer cells with high expression of the M3 were used as an in vitro model,and 22 samples were tested with a firefly luciferase reporter gene assay system using a Ca2+response element?NFAT-RE?as a specific promoter.3.The"wind-heat syndrome"treated by SFJD was equivalent to the upper respiratory tract infection of modern medicine,and the bitterness can remove heat and cure wind-heat syndrome,which is related to its anti-inflammatory effect.Therefore,the macrophage?RAW264.7?induced by lipopolysaccharide?LPS?was used as an inflammatory model and was randomly divided into positive drug group?dexamethasone?,blank group,model group and 22 sample administration groups?n=3?.ELISA method was used to detect TNF-?and IL-6 in cell supernatant.4.With the relative peak area of 44 components in 22 samples as independent variables,the activation rate of M3 receptor and the inhibition rate of LPS induced IL-6 and TNF-alpha inflammatory factors were the dependent variables.Matlab 7.13combined with GA toolbox for model research,the use of BP-neural network algorithm,and the number of neurons in the model and training algorithm to screen and contrast research,the use of genetic algorithm to optimize the model parameters,set up a spectral effect relationship model,"spectrum"and"efficiency"to carry on the correlation analysis.Result:The peak extraction,peak-matching,and PCA analysis were perfor med on the positive and negative mode data of 22 samples using Markerlynx software.The relative peak areas of 44 characteristic peaks were obtained as t he independent variable for subsequent spectral-effect analysis.Acquiring data on the efficacy and anti-inflammatory efficacy data of 22 samples with differen t ratios of detoxification and detoxification on M3 receptors.Through the estab lishment of the best artificial neural network model,16 characteristic peaks wer e found to have the greatest influence on the activity of M3 receptor,and the14 characteristic peaks were closely related to the anti-inflammatory activity,which may be the effective components of SFJD.Conclusion:Based on the trilism of"property-response-component"and the"spectrum-effect"association analysis,this paper has preliminarily clarifies the material basis of the SFJD"sparse wind solution table"and"heat-clear and t oxin-resolving".This study lays a foundation for the study of“drug property-E ffectiveness-Substance Basics”of SFJD and provides evidence for the determin ation of its Q-Marker.
