Effect Of Tongmai Quzhuo Decoction On Anti-inflammatory And Analgesic Effects And Its Effect On Inflammatory Factors In Rat Models Of Gouty Arthritis

Objective To explore the effect of Tongmai Quzhuo Decoction on anti-inflammatory and analgesic effects and its influence on inflammatory factors in rats with gouty arthritis.Methods 1.260 Kunming mice were randomly divided into normal control group,Lijia Lixian group,Tongfengding capsule group,high-dose Tongmai Quzhuo Decoction group,Tongmai Quzhuo Decoction group,Tongmai Quzhuo Decoction low-dose group,10 in each group.The ear swelling caused by xylene,paw swelling caused by egg white,writhing experiment,and hot plate experiment were used to observe the anti-inflammatory and analgesic effects of Tongmai Quzhuo Decoction.2.84 Wistar rats,male,were randomly divided into normal group,model group,high-dose Tongmai Quzhuo Decoction group,Tongmai Quzhuo Decoction group,Tongmai Quzhuo Decoction low-dose group,Tongfengding capsule group,Li Galician group.12 in each group,after screening,10 for each group and observe the general changes.The content of uric acid was detected by automatic biochemical analyzer.The levels of IL-1? and TNF-? in synovium of rats were detected by ELISA.The XOD activities in serum and liver were detected by xanthine oxidase kit.The results were analyzed by Real-Time qPCR.Methods The expression of IL-1? and TNF-? in synovium of rats was detected.Rat synovial membranes were taken for pathological examination,and histopathological morphological changes were observed under HE light microscope.Results 1.Tongmai Quzhuo Decoction has obvious anti-inflammatory and analgesic effects and showed a certain dose-effect relationship.Among them,Tongmai Quzhuo Decoction high-dose group had the best effect,followed by Tongmai QuzhuoDecoction middle dose group.2.Tongmai Quzhuo decoction each dose group can reduce the degree of uric acid(UA)value of rats with gouty arthritis in different degrees,improve the pathological morphology of the joints of rats in the administration group;Tongmai Quzhuo Decoction each dose group It can inhibit the activity of xanthine oxidase(XOD)in rat serum and liver,and regulate the contents of IL-1? and TNF-? and the expression of IL-1? and TNF-? mRNA in rat joint soft tissue and synovial fluid.The effect of the high dose group of pulse Quzhuo Decoction was the most obvious,followed by Tongmai Quzhuo Decoction group.Conclusion 1.Tongmai Quzhuo Decoction has obvious anti-inflammatory and analgesic effects,and shows a certain dose-effect relationship,especially for high-dose Tongmai Quzhuo Decoction.2.Tongmai Quzhuo Decoction can significantly improve the diet and joint activity of rats,reduce the serum UA value and related pathological parameters of rats,and at the same time can inhibit XOD activity in serum and liver of model rats,and regulate rat joint soft tissue and Synovial fluid IL-1?,TNF-? content and IL-1?,TNF-? mRNA expression,and then improve the condition of gouty arthritis.The results showed a certain dose-effect relationship between the doses of Tongmai Quzhuo Decoction.
